Glucose-dependent trans-plasma membrane electron transport and p70S6k phosphorylation in skeletal muscle cells

Abstract: The reduction of extracellular oxidants by intracellular electrons is known as trans-plasma membrane electron transport (tPMET). The goal of this study was to characterize a role of tPMET in the sensing of glucose as a physiological signal. tPMET from C2C12 myotubes was monitored using a cell-impermeable extracellular electron acceptor, water-soluble tetrazolium salt-1 (WST-1). Superoxide dismutase in the incubation medium or exposure to an NADPH oxidase (NOX) isoform 1/4 inhibitor suppressed WST-1 reduction b… Show more

“…Impaired mitochondrial metabolism is followed by ROS production and inflammation. Several mechanisms have been proposed to explain impaired mitochondrial metabolism, such as mitochondrial complex inactivation and mitochondrial metabolism switch from oxidative phosphorylation to glycolysis [ 35 , 36 ]. It is necessary to explore whether TGP has the ability to normalize mitochondrial metabolism in cardiomyocytes.…”

“…A small-molecule inhibitor of vascular permeability and angiogenesis interacts with MEK1 and suppresses a pERK-FosB/ΔFosB-VCAM-1 axis ( Li et al, 2020 ). P70 S6K is the first signal transduction element inhibited by mTOR in mammalian cells, as well as a downstream effector of mTORC1 ( Kelly et al, 2019 ); it participates in many physiological processes, including cell survival, protein synthesis, and mRNA splicing, by phosphorylating various substrates ( Tetsuo et al, 2019 ). S6K1 absence leads to hypoinsulinemia, glucose intolerance, and β-cell disorder ( Um et al, 2004 ).…”
