Glucose-dependent phosphorylation signaling pathways and crosstalk to mitochondrial respiration in insulin secreting cells

Abstract: Background Glucose is the main secretagogue of pancreatic beta-cells. Uptake and metabolism of the nutrient stimulates the beta-cell to release the blood glucose lowering hormone insulin. This metabolic activation is associated with a pronounced increase in mitochondrial respiration. Glucose stimulation also initiates a number of signal transduction pathways for the coordinated regulation of multiple biological processes required for insulin secretion. Methods Shotgun p… Show more

“…We considered only genes with P ≤ 0.05 and found a set of 208 genes, of which 41 and 8 were, respectively, upregulated and downregulated in all three datasets ( Supplementary Table S4 ). Interestingly, several of such genes (20 upregulated and 4 downregulated) have been reported to be linked to diabetes ( 74 , 76 , 91–93 , 111 , 112 , 126–138 ) ( Supplementary Table S4 ).…”

“…Akt is a protein kinase, an important member of the insulin signaling pathway. It also regulates various lipid biosynthesis enzymes such as fatty acid synthase (FASN), ATP-citrate lyase (ACLY), and acetyl CoA carboxylase (ACC) through SREBP-1 [ 52 ]. Amongst the three isoforms of SREBP (SREBP-1a, SREBP-2, and SREBP-1c), SREBP-1c and SREBP-1a are mostly involved in fatty acid biosynthesis, and SREBP-2 is involved in cholesterol biosynthesis.…”

“…Alternatively, PKC-1 regulate mitochondrial function. Calcium-independent PKCs interact with a number of mitochondrial proteins 157 , and have been proposed to promote insulin secretion by regulating mitochondrial ATP production in pancreatic beta cells 177,178 . Finally, PKC-1 may regulate DCV secretion through a less direct mechanism.…”
