Glucose Clamping Using the Biostator GCIIS

Abstract: The Biostator GCIIS was used to clamp circulating glucose at hypoglycemic (42 +/- 1 mg/dl), euglycemic (86 +/- 2 mg/dl), and hyperglycemic (142 +/- 1 mg/dl) levels in normal subjects during a concomitant infusion of insulin (0.1 U/kg/h). Because of limitations in maximal glucose infusion (1 g/min) from the Biostator, a supplementary infusion of glucose was required to accomplish euglycemic and hyperglycemic clamps. The coefficients of variation of blood glucose were 7.06 +/- 1.3%, 5.9 +/- 0.5%, and 6.1 +/- 0.9… Show more

“…For the Biostator a mean CV of the BG concentration of 5.9 ± 0.5% was observed in a euglycemic clamp study in healthy people. 24 The present 39 ClampArt clamps resulted in a slightly lower CV at the target level of 5.5 ± 2.1%. c. Accuracy (as well as trueness and precision) refers to the quality of the measurement, not to the quality of control.…”

“…The improve ment in tissue insulin sensitivity demonstrated using the clamp technique may explain the evolution of the insulin need. This technique is now routinely used and results are reproducible [7,8]. We demonstrated for each steady state o f the clamp an improvement in glucose consump tion for similar hyperinsulin levels.…”

“…Tissue insulin sensitivity was measured by the glucose clamp technique as described by De Fronzo et al [6] and modified by Verdonk et al [7] and us [8]. The test was performed before begin ning the dietary treatment and after 3 months o f diet.…”

“…Along with insulin infusion and to avoid hypoglycemia, variable amounts of glucose were infused according to the principles of the euglycemic glucose clamp method first described by De Fronzo et al [6] and applied to the Biostator by Verdonk et al [7], The rate of glucose infusion was automatically adjusted by algorithms of the Biostator. Glucose was infused as a 30% solution to which 0.26 mEq KCl/ml was added to prevent hypokalemia.…”
