Glucose and Insulin Induction of Bile Acid Synthesis

Abstract: Background: Bile acid synthesis plays an important role in nutrient absorption and maintaining metabolic homeostasis under normal physiology. Results: Glucose induces cholesterol 7␣-hydroxylase activity and postprandial bile acid synthesis via insulin signaling and epigenetic mechanisms. Conclusion: Glucose and insulin are major postprandial factors that stimulate bile acid synthesis to maintain hepatic metabolic homeostasis. Significance: Nutrient regulation of bile acid synthesis is impaired in diabetes and … Show more

“…Furthermore, oral glucose ingestion leads to increased levels of cholecystokinin, a hormone that stimulates the production of hepatic bile and gall bladder contractions (Liddle et al 1985). The link between bile acids and glucose homoeostasis was further confirmed by the demonstration of enhanced Cyp7a1 mRNA expression following insulin injection or oral glucose administration in fasting mice (Li et al 2012). Additionally, in primary human hepatocytes, insulin and glucose were shown to stimulate CYP7A1 mRNA expression (Li et al 2006(Li et al , 2010b, which suggests the existence of glucose/insulin-regulated gene transcription in the liver.…”

“…These results support an important role of FGF15, synthesized in the intestines by activation of FXR, in the regulation of CYP7A1 and bile acid synthesis in the liver. CYP7A1 has also been reported to be down-regulated by glucagon (24, 25) and pro-inflammatory cytokines (15) and up-regulated by glucose and insulin during the postprandial period (26). …”
