Glucosamine Long-Term Treatment and the Progression of Knee Osteoarthritis: Systematic Review of Randomized Controlled Trials

Abstract: The available evidence suggests that glucosamine sulfate may be effective and safe in delaying the progression and improving the symptoms of knee OA. Due to the sparse data on structural efficacy and safety, further studies are warranted.

“…6B), suggesting the involvement of post-ER degradation under conditions in which N-linked glycosylation is not blocked and proteasomal degradation is inhibited/minimized. Glucosamine, widely synthesized endogenously and also sold commercially for the treatment of arthritis (28,29), has recently been linked, controversially, to insulin resistance in both cultured cells and animal models (30,31). Increased plasma levels of apoB-100-containing lipoproteins are an important risk factor in the development of atherosclerosis and are commonly found in insulinresistant states and type 2 diabetes (32).…”

“…In addition, middle ear admittance and hearing characteristics should also be mon-itored in patients who are taking drugs such as glucosamine sulfate and diacerein that may slow the degeneration of cartilage [Sarzi-Puttini et al, 2005;Poolsup et al, 2005]. Middle ear surgical intervention has been suggested for selected patients with rheumatoid arthritis [Salvinelli et al, 2004].…”
