Glucocorticoid receptor expression in inflammatory bowel disease: evidence for a mucosal down‐regulation in steroid‐unresponsive ulcerative colitis

Abstract: Summary Background : Glucocorticoids (GC) play a major role in the attenuation of inflammation. Glucocorticoid receptor (GR) expression is an important determinant of steroid sensitivity. Aims : To investigate whether GR mRNA expression is altered in inflammatory bowel disease, and whether GR mRNA expression correlates with disease activity and may predict response to GC therapy. Methods : Mucosal biopsies were taken from 33 patients with ulcerative colitis, 21 with Crohn's disease and 11 controls. Peripheral … Show more

“…Currently, we do not have any useful marker for predicting the future response to LCAP, comparable the expression of the glucocorticoid receptor on leukocytes or the local mucosa, which has been suggested as a possible marker for determining the steroid response. 25 Our results show that the clinical response to LCAP was associated with modulations of population and/or platelet functions, especially after the first LCAP.…”

“…Quantitative reverse transcription polymerase chain reaction (RT-PCR) was performed by real-time PCR as described earlier. 19 Primer pairs for H ϩ /K ϩ -ATPase and ␤-actin are summarized in the Table. Each sample was analyzed in duplicate. The investigated number of cDNA molecules was related to ␤-actin cDNA molecules detected in the same sample to normalize for the quantity of RNA extracted and the efficiency of cDNA synthesis.…”

“…The relative expression was then calculated as described. 19 Developed Tension (mN/mm²) (Figures 1 and 4), or the Tukey test was used ( Figure 6). The authors had full access to and take full responsibility for the integrity of the data.…”

“…Although the leukocyte recruitment by adhesion molecules plays a role in the initiation and progression of the disease, there is little documentation of their roles as biomarkers. The gene expression of ICAM-1 is elevated in UC but was not correlated with response to steroids (Raddatz et al, 2004). In the same study low expression of the glucocorticoid receptor was correlated to non-response.…”
