Background Chronic constipation is a gastrointestinal functional disorder which affects patient quality of life. Therefore, many studies were oriented to search herbal laxative agents. In this study, we investigated the effect of Globularia alypum L. leaves aqueous extract (GAAE) against loperamide (LOP)‐produced constipation. Methods Animals were given LOP (3 mg/kg, b.w., i.p.) and GAAE (100, 200, and 400 mg/kg, b.w., p.o.) or yohimbine (2 mg/kg, b.w., i.p.), simultaneously, for 1 week. Gastric‐emptying test and intestinal transit were determined. Colon histology was examined, and oxidative status was evaluated using biochemical‐colorimetric methods. Key Results GAAE ameliorates significantly gastric emptying (64% to 76.5%) and intestinal transit (66.65% to 84.73%). LOP negatively influenced defecation parameters and generated a stress situation. GAAE administration in contrast ameliorated those parameters and re‐established oxidative balance. Conclusion GAAE showed a modest action against oxidative stress and decreased LOP effect and thereby can be considered a pharmacological agent in constipation.

Introduction In the quest for new sources of biologically‐active compounds, the chemical, and biological profiles of two Globularia species (G. trichosantha Fisch. & C. A. Mey and G. orientalis L.) were investigated. Methodology Chemical profiles were evaluated by high‐performance liquid chromatography coupled to electrospray ionisation and quadrupole time‐of‐flight mass spectrometry (HPLC‐ESI‐QTOF‐MS), as well as by their total phenolic, flavonoids, and phenolic acids contents. The antioxidant abilities of the investigated extracts were done using different assays including free radical scavenging [1,1‐diphenyl‐2‐picryl‐hydrazyl (DPPH) and 2,2′‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulphonic acid (ABTS)], reducing power (cupric reducing antioxidant capacity and ferric reducing antioxidant power), phosphomolybdenum, and metal chelating. Inhibitory potential against key enzymes involved in neurodegenerative diseases (cholinesterases; AChE, and BChE), diabetes (α‐glucosidase and α‐amylase), hyperpigmentation (tyrosinase) and obesity (pancreatic lipase) were evaluated. Results Globularia trichosantha and G. orientalis extracts showed remarkable antioxidant properties, with the water extracts being a better source of antioxidant compounds. Both species showed remarkable inhibitory effects against the target enzymes. However, for both species, only the acetyl acetate and methanolic extracts were potent against cholinesterases and lipase. Conclusion HPLC‐ESI‐QTOF‐MS analysis revealed the presence of 107 compounds from G. trichosantha and G. orientalis, among which, 43 compounds have been preliminarily characterised for the first time from the Globulariaceae family. To date, this study can be considered as the most comprehensive research focused on the characterisation of G. trichosantha and G. orientalis. Results amassed from this study tend to show that these plants represent a rich source of biologically active compounds which can be further explored and validated for their therapeutic potential.

This is the first study to evaluate the topical application of Lobelia alsinoides Lam (LT) ethanolic extract on burns in rats. A deep third-degree burn wound was inflicted in adult male Wistar rats and the burns were dressed daily with a topical ointment formulation (Patent filed) comprising of Lobelia alsinoides Lam (5% and 10% w/w). The wound had noteworthy contraction and quicker eschar removal in 10% w/w LTtreated groups followed by 5% w/w treated groups on comparing with the commonly prescribed ointment (Silverex TM containing 1% w/w Silver sulfadiazine). Histopathological analysis showed that ointment containing 10% w/w LT ethanolic extract significantly increased fibroblast growth, which plays a major role in anatomic integrity, collagen synthesis, and accelerated the rate of the healing process. This study shows that the ethanolic extract of Lobelia alsinoides Lam, a previously pharmacologically unreported traditional medicinal plant, possesses wound contraction and eschar removal properties on burn wounds.
