ABSTRACT. Herbal preparations of Globimetula braunii leaves are used in tropical African countries for the treatment of several illnesses. A preliminary study of the leaf extracts of Globimetula braunii has confirmed the antioxidant, antibacterial, biochemical and toxicological potency of the plant. The leaf extracts were subjected to bioactivity guided fractionation that led to the isolation of the active compounds. Antioxidant, antityrosinase and antimicrobial inhibitory activities of the isolated compounds were evaluated. Structural elucidations of the isolated compounds were carried out spectroscopically, including the use of 1D/2D nuclear magnetic resonance (NMR), Fourier transform infrared (FTIR) spectroscopies and mass spectrometry. Results indicated that quercetin (15) isolated from ethyl acetate fraction exhibited the highest 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) scavenging capacity and also showed the highest ferric reducing antioxidant potential (FRAP) equivalent value. Quercetin (15) and dodoneine (14) displayed the highest tyrosinase inhibition activity with IC50 value of 0.12±0.32 mM and 0.19±0.20 mM, respectively. All the isolated compounds showed weak to not active in the antimicrobial inhibition assay. The findings in this report have revealed the antioxidant and antityrosinase potential of Globimetula braunii and thus supporting their traditional uses. Ganoderma lucidum triterpenoids (GLT) possess advantageous characteristics including antiinflammatory, anti-oxidant and anti-cancer properties. In this study, a drug loaded core-shell fibrous system was engineered using the coaxial electrospinning method. The core comprised polycaprolactone and GTL whilst the shell contained polyethylene oxide with anti-cancer drug methotrexate. The morphology and structure, chemical composition, biocompatibility and in vitro release profile of the encapsulated GLT was assessed. Scanning electron microscopy images showed cylindrical fibers were developed whilst fluorescence microscopy confirmed the successful formation of core-shell fibers.Increasing GLT concentration from 2 wt% to 6 wt% resulted in an increase in mean fiber diameter from 2.29 ± 1.26 µm to 4.65 ± 2.30 µm, respectively. Fourier transform infrared spectroscopy, X-ray diffraction and differential scanning calorimetry analysis all confirmed that both drugs were encapsulated in amorphous form while contact angle hysteresis showed engineered fibres to possess good hydrophilicity. In vitro drug release tests indicated the release of methotrexate (R 2 = 0.9941) and GLT (R 2 = 0.9924) were controlled by Fick Diffusion. In vitro cell culture experiments proved that the combined use of traditional Chinese medicine and current western medicine has good application prospects in the treatment of cervical cancer.

Globimetula braunii is a hemi-parasitic plant used in African ethnomedicine for the management of microbial infections, rheumatic pain and tumors amongst others. We report the isolation and characterization of eight compounds with their antioxidant and antimicrobial activities. The air-dried powdered leaf was macerated in EtOH/H20 (4:1). The extract was solvent-partitioned into n-hexane, EtOAc, n-BuOH and aqueous fractions. The fractions were screened for their antioxidant properties, using DPPH, FRAP, TAC and FIC assays. Antimicrobial analysis was performed using the micro-broth dilution method. The active EtOAc fraction was purified for its putative compounds on a repeated silica gel column chromatography monitored with TLC-bioautography. The isolated compounds were characterized using spectroscopic methods of UV, FT-IR, NMR and MS. Eight compounds (1–8) were isolated and characterized as 13,27-cycloursane (1), phyllanthone (2), globraunone (3), three phenolics: methyl 3,5-dihydroxy-4-methoxybenzoate (4), methyl 3-methyl-4-hydroxybenzoate (5) and guaiacol (6), as well as two phenol derivatives: 4-formaldehyde phenone (7) and 6-methoxy-2H-inden-5-ol (8). The study identified 4 and 6 as natural antioxidant compounds with potential as antimicrobial agents.
