-globin mRNAs capped with m7G, m22.7G or m32.2.7G differ in intrinsic translation efficiency

Abstract: We examined the intramolecular effect of altered cap structures on translation efficiency of artificial beta-globin mRNAs. For these studies, synthetic dinucleotides of the form X(5')ppp(5')G [X = 7-methyl guanosine (m7G), 2,7-dimethyl guanosine (m2(2,7)G) or 2,2,7-trimethyl guanosine (m3(2,2,7)G)], were transcriptionally incorporated into mRNAs, containing rabbit beta-globin coding sequences, using T7 RNA polymerase and a beta-globin cDNA template. These synthetic mRNAs were assayed in reticulocyte lysate for… Show more

“…There is some concern that if an RNA has a TMG cap instead of an m7G cap, this could have repercussions for the initiation of translation. The proportion of hypermethylated guanosine-capped RNAs was found to be lower in polysomal RNA than in nonpolysomal RNA, suggesting that TMG RNAs may not be translated efficiently (57,58). How these in vitro findings reflect in vivo translation is not entirely clear, however.…”

“…Gp 3 G are translated in a rabbit reticulocyte system 1.5-fold more efficiently than m 7 Gp 3 G-capped mRNAs (Darzynkiewicz et al 1988). Similarly, mRNAs containing the bulky benzyl group on N7 (bn 7 Gp 3 G) instead of methyl are translated 1.7-fold more efficiently than mRNAs capped with the natural structure (Darzynkiewicz et al 1989).…”

“…As shown in Figure 2B, trans-spliced products are present in both the supernatant and the pellet. The poor precipitation of the trans-spliced product is likely attributable to the fact that it contains a trimethylguanosine cap (Liou et al 1990) that binds less efficiently to mammalian eIF4E (Darzynkiewicz et al 1988). In contrast, Ur-RNA is not detected at all in the pellet, suggesting it does not contain a 5Ј cap.…”
