Enhanced biological phosphorus removal (EBPR) is strongly influenced by the influent ratio of readily biodegradable carbon to soluble phosphorus due to the preferences of phosphorus-accumulating organisms (PAO). The sidestream EBPR (S2EBPR) process redirects a portion of return activated sludge (RAS) to a sidestream fermenter, increasing the availability of biodegradable carbon. In this study, we assessed the performance and microbial community structure of a full-scale S2EBPR demonstration supplemented with external carbon dosing. By the end of the 10 month study period, the demonstration achieved a median effluent orthophosphate of 0.3 mg/L. The microbial community consisted of a common core microbiome in the RAS fermenter, EBPR basin, and nitrification basin. The most abundant PAO detected were Ca. “Dechloromonas phosphorivorans”, while the canonical PAO Ca. “Accumulibacter” and Tetrasphaera were observed in lower relative abundance. In addition to non-canonical PAO enrichment, the glycogen-accumulating organism Ca. “Competibacter” proliferated throughout the study and at some points outnumbered PAO taxa by 30 to 1 with no tangible performance impacts. This study provides insights into successful S2EBPR implementation at a low-carbon facility and improves our understanding of microbial community structure and key PAO and GAO in S2EBPR systems.

Candidatus Accumulibacter is a key genus of polyphosphate-accumulating organisms (PAOs) found in laboratory- and full-scale wastewater treatment systems, mediating enhanced biological phosphorus removal (EBPR). However, little is known about their ability to resist phage infection. We conducted a systematic analysis of the occurrence and characteristics of clustered regularly interspaced short palindromic repeats and associated proteins (CRISPR-Cas) systems and prophages in diverse Ca. Accumulibacter taxa (43 in total, including 10 newly recovered genomes). Fourty complete CRISPR loci were identified in 28 genomes, falling into 6 subtypes. The occurrence and distribution of CRISPR-Cas systems did not follow a vertical evolutionary relationship. Phylogenetic analyses of the cas genes and direct repeats (DRs) suggested that the CRISPR-Cas systems were likely acquired via horizontal gene transfer, with acquisition rates higher than those of speciation, rendering different Ca. Accumulibacter distinct adaptivity to phage predations. 2448 spacers were identified, 67 of them matched to known phages. Major differences were observed among the numbers of spacers for different Ca. Accumulibacter, showing unique phages that could be resisted by different members. A comparison of the spacers in genomes having the same cas gene but from distinct geographical locations indicated that habitat isolation may have resulted in the acquisition of different spacers by different Ca. Accumulibacter. Metagenomic analysis allowed the identification of 26 viral contigs (18 are Caudovirales members) in 6 metagenomic datasets from three lab-scale enrichment reactors, matching to 73 spacers in 10 Ca. Accumulibacter genomes in these reactors, showing the specific immunity of these Ca. Accumulibacter. Metatranscriptomic analyses showed the activity of the CRISPR-Cas system under both anaerobic and aerobic conditions. Extra efforts were made to identify prophages in Ca. Accumulibacter genomes. In total, 133 prophage regions were identified. Twenty-seven of them were classified as potentially active prophages. Three prophages (all are Caudovirales members, in DS2011, SCELSE-7IIH and SCELSE-5IIH, respectively) are readily activable. Differences in the occurrence of CRISPR-Cas systems and prophages in Ca. Accumulibacter will lead to their distinct responses under phage predation. This study represents the first systematic analysis of CRISPR-Cas systems and prophages with combined experimental and bioinformatic methods in the Ca. Accumulibacter lineage, providing new perspectives on the potential impacts of phages on Ca. Accumulibacter and EBPR systems.

Phosphorous (P) removal in wastewater treatment is essential to prevent eutrophication in water bodies. Side‐stream enhanced biological phosphorous removal (S2EBPR) is utilized to improve biological P removal by recirculating internal streams within a side‐stream reactor to generate biodegradable carbon (C) for polyphosphate accumulating organisms (PAOs). In this study, a full‐scale S2EBPR system in a water resource recovery facility (WRRF) was evaluated for 5 months. Batch experiments revealed a strong positive correlation (r = 0.91) between temperature and C consumption rate (3.56–8.18 mg‐COD/g‐VSS/h) in the system, with temperature ranging from 14°C to 18°C. The anaerobic P‐release to COD‐uptake ratio decreased from 0.93 to 0.25 mg‐P/mg‐COD as the temperature increased, suggesting competition between PAOs and other C‐consumers, such as heterotrophic microorganisms, to uptake bioavailable C. Microbial community analysis did not show a strong relationship between abundance and activity of PAO in the tested WRRF. An assessment of the economic feasibility was performed to compare the costs and benefits of a full scale WRRF with and without implementation of the S2EBPR technology. The results showed the higher capital costs required for S2EBPR were estimated to be compensated after 5 and 11 years of operation, respectively, compared to chemical precipitation and conventional EBPR. The results from this study can assist in the decision‐making process for upgrading a conventional EBPR or chemical P removal process to S2EBPR.Practitioner Points Implementation of S2EBPR presents adaptable configurations, exhibiting advantages over conventional setups in addressing prevalent challenges associated with phosphorous removal. A full‐scale S2EBPR WRRF was monitored over 5 months, and activity tests were used to measure the kinetic parameters. The seasonal changes impact the kinetic parameters of PAOs in the S2EBPR process, with elevated temperatures raising the carbon demand. PAOs abundance showed no strong correlation with their activity in the full‐scale S2EBPR process in the tested WRRF. Feasibility assessment shows that the benefits from S2EBPR operation can offset upgrading costs from conventional BPR or chemical precipitation.
