Global Modulation in DNA Epigenetics during Pro-Inflammatory Macrophage Activation

Abstract: DNA methylation patterns create distinct gene expression profiles. These patterns are maintained after cell division, thus enabling the differentiation and maintenance of multiple cell types from the same genome sequence. The advantage of this mechanism for transcriptional control is that chemical-encoding allows to rapidly establish new epigenetic patterns "on-demand" through enzymatic methylation and de-methylation of DNA. Here we show that this feature is associated with the fast response of macrophages dur… Show more

“…The epigenetic level of regulation is critical for the differentiation and functional programming of macrophages [ 15 , 113 ]. There are three levels of epigenetic control of macrophages differentiation and activation: DNA methylation, histone modifications, and microRNA [ 15 , 114 ].…”

“…TET2 expression is increased in intratumoral myeloid cells, both in a mouse model of melanoma and in melanoma patients, that is dependent on an IL-1R-MyD88 pathway [ 135 ]. Recently, the combination of mass spectrometry and single molecular imaging demonstrated that LPS induces global changes in DNA methylation of the genome of murine macrophages [ 113 , 134 ].…”

“…Furthermore, these results by sequence analysis motivated us to determine the biological function of porcine DNMT3B. Given the reported data that expression of DNMT3B is downregulated in LPS-activated mouse macrophages, which is associated with the global demethylation of DNA [ 28 ], thus we chose the LPS-activated PAM as our model for the functional analysis of porcine DNMT3B. Since the alteration of DNMT3B in the LPS-stimulated porcine alveolar macrophages remains unknown, we first investigated the expression profile of DNMT3B in the LPS-activated PAM.…”
