Global expression profiling of yeast treated with an inhibitor of amino acid biosynthesis, sulfometuron methyl

Abstract: The expression pattern of 1,529 yeast genes in response to sulfometuron methyl (SM) was analyzed by DNA microarray technology. SM, a potent herbicide, inhibits acetolactate synthase, a branched-chain amino acid biosynthetic enzyme. Exposure of yeast cells to 0.2 microg/ml SM resulted in 40% growth inhibition, a Gcn4p-mediated induction of genes involved in amino acid and cofactor biosynthesis, and starvation response. The accumulation of intermediates led to the induction of stress response genes and the repre… Show more

“…6). SMM is a specific inhibitor of acetolactate synthase, which causes a depletion of branched-chain amino acids in the cell (Jia et al 2000). Surprisingly, the nab4 mutants showed a growth advantage in comparison to wild-type cells when exposed to SMM.…”

“…Furthermore, in the absence of any amino acid deficiency, a basal level of Gcn4 mRNA translation is required for the transcription of several genes (e.g., HIS3, ARG4, ARG3, and ARO3) (57). Two genomic studies have aimed to identify the whole set of GAAC target genes (68,90). In one of them (90), 539 genes whose expression is activated in a Gcn4-dependent manner in amino acid-starved cells were listed.…”

“…This system mediates increased transcription of more than 500 genes in response to starvation for an amino acid (2)(3)(4). The induced genes encode amino acid and purine biosynthetic enzymes and a variety of proteins involved peripherally in amino acid biosynthesis or in amino acid uptake or salvage (autophagy) (4,5).…”
