Global Characterization of Protein Secretion from Human Macrophages Following Non-canonical Caspase-4/5 Inflammasome Activation

Abstract: Gram-negative bacteria are associated with a wide spectrum of infectious diseases in humans. Inflammasomes are cytosolic protein complexes that are assembled when the cell encounters pathogens or other harmful agents. The non-canonical caspase-4/5 inflammasome is activated by Gram-negative bacteria-derived lipopolysaccharide (LPS) and by endogenous oxidized phospholipids. Protein secretion is a critical component of the innate immune response. Here, we have used label-free quantitative proteomics to characteri… Show more

“…Newly synthesized proteins are targeted for entry into secretory pathways by the presence of an N-terminal signal sequence (Lorey, Rossi, Eklund, Nyman, & Matikainen, 2017;Martoglio & Dobberstein, 1998). These proteins transmit and receive information crucial to the health and adaptive ability of organisms (Walter & Ron, 2011).…”

“…On the other hand, cleaved GsdmD can promote Nlrp3‐dependent activation of caspase‐1 and IL‐1β secretion against Gram‐negative bacterial infection 142 . A third effector function involves anti‐microbial protein secretion, as activation of non‐canonical caspase‐4/5 inflammasome results in secretion of multiple proteins by human primary macrophages, including S100A8 and prothymosin‐α, which represent TLR4 ligands, 149 while the exact contribution of these secreted proteins in host defense against bacterial invaders remains to be investigated.…”

“…The tryptic peptides were concentrated and purified using ZipTip C18 (Millipore) and equivalent quantities of peptides from each sample were submitted to an Easy-nLC 1000 nano-LC system (Thermo Scientific,) coupled with a quadrupole Orbitrap mass spectrometer (Q ExactiveTM, ThermoElectron, Bremen, Germany) equipped with a nanoelectrospray ion source (Easy-SprayTM, Thermo Scientific), as previously reported [16]. The liquid chromatography (LC) separation was performed in an Easy-SprayTM column capillary with a 25-cm bed length (C18, 2-µm beads, 100 Å, 75-µm inner diameter, Thermo Scientific), using a flow rate of 300 nl/min.…”
