Glial fibrillary acidic protein in the retina of the developing albino rat: An immunoperoxidase study of paraffin‐embedded tissue

Abstract: The peroxidase-anti-peroxidase method was used on paraffin-embedded material to demonstrate the distribution of glial fibrillary acidic (GFA) protein, an astrocyte-specific protein, in the developing retina of the albino rat. At birth activity was scant and was confined to scattered, poorly differentiated cells in the inner retinal layers near the optic disc. At 3 days primitive astrocytes which displayed GFA protein activity were confined to the stratum opticum near the optic disc. With increasing age these c… Show more

“…Immunostaining of retinal wholemounts using antibodies directed against GFAP confirmed previous findings regarding the location and general morphology of rat retinal astrocytes (Dixon and Eng, 1981;Stone and Dreher, 1987;Chan-Ling and Stone, 1991). GFAP-IR astrocytes were found throughout the nerve fiber layer of the rat retina.…”

“…This observation is consistent with our impression that GFAP-IR processes do not extend completely underneath veins. GFAP-IR processes were observed to follow capillaries as far as the inner plexiform layer, as observed previously in the retinas of albino rats (Dixon and Eng, 1981) and humans (Triviñ o et al, 1997).…”

“…Identical hybridization patterns are observed with both partially digested and full-length RNA probes. Labeling is not associated with endothelial cells lining blood vessels nor with astrocytes, which are mainly located in the ganglion cell axon layer (Dixon and Eng, 1981;Schnitzer, 1988). Only background labeling is observed over the distal INL, outer plexiform layer and outer nuclear layer, and in sections incubated with the sense RNA probe (Fig.…”

“…Ethanol fixation improved the staining intensity of OF AP in gray matter astrocytes compared with that obtained with formalin or aldehyde fixatives (Dixon and Eng, 1981) and allowed astrocyte immunostaining of vimentin (personal observations). Pri mary antibodies, diluted I: 100, included rabbit anti OFAP polyclonal IgO (Dako Corp.), mouse monoclonal anti-OFAP IgO (Oncogene Science, Inc.), and mouse monoclonal anti-vimentin IgO (Dako Corp.).…”
