Ginsenoside Rg2 Ameliorating CDAHFD-Induced Hepatic Fibrosis by Regulating AKT/mTOR-Mediated Autophagy

Abstract: Ginsenoside Rg2 (G-Rg2) in the rhizome of Panax ginseng can modify lipid accumulation, oxidative stress, and apoptosis in the liver induced by a high-fat diet. This research adds to this by assessing the potential antifibrosis effect of G-Rg2 (including possible mechanisms). G-Rg2 significantly improved pathological changes in liver tissue induced by a choline-deficient, l-amino acid-defined, high-fat diet (CDAHFD), it inhibited serum transaminase, plasma lipopolysaccharide, and liver hydroxyproline levels; it… Show more

“…Activated AKT could phosphorylate mTOR, promoting the conversion of lung epithelial cells to fibroblasts, inducing lung fibrosis [ 40 ]. Additionally, the activation of AKT/mTOR pathway also gives rise to oxidative stress, which further aggravated the development of fibrosis [ 29 , 41 ]. Here, our study unveiled that EG treatment not only decreased oxidative stress but also inhibited EMT during PM2.5-induced lung fibrosis by blocking the activation of AKT/mTOR pathway.…”

“…Changes in the mRNA levels of each gene were quantified using qRT-PCR [ 13 ]. Briefly, total hepatic RNA was extracted using TRIzol reagent, and the purity and concentration of the RNA were determined using a NanoDrop 2000C spectrophotometer (Thermo Scientific, Waltham, MA USA).…”

“…Ginsenoside Rg2 (G-Rg2) reinstated the impaired oleic acid-induced autophagic flux and inhibited TGF-β1 expression by promoting p62 degradation in hepatocytes. Mechanistically, G-Rg2 inverted the activation and inhibited the autophagy of HSCs through the AKT/mTOR signaling pathway [211] .…”
