Gingival crevicular fluid matrix metalloproteinase‐13 levels and molecular forms in various types of periodontal diseases

Abstract: These results indicate that elevated GCF MMP-13 levels may play an important role in the pathogenesis of CP. These data demonstrate, for the first time, pathologically activated and elevated MMP-13 in GCF.

“…Total RNA isolated from tendons or cultured tenocytes (RNeasy Kit, Eppendorf, Westbury, NY, USA) was reverse transcribed with MMLV reverse transcriptase and an Oligo(dT) [12][13][14][15][16][17][18] primer and expression of MMP-13 was quantitated by real time PCR (ABI Prism 7900HT RealTime PCR System, Applied Biosystems, Framingham, MA, USA). Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was used for normalization.…”

“…Furthermore, because IL-1b induces MMP-13 [10,18,39,42], we hypothesized changes in tenocyte MMP-13 expression would be dependent on expression of IL-1b.…”

“…Despite MMP-13 has been more related to chronic periodontitis progression than severity of inflammation, it has previously been reported that MMP-13 also increase progressively from healthy subjects to periodontitis in GCF. Higher levels of active forms and total MMP-13 have been described in gingivitis versus healthy sites; whereas MMP-13 levels and active forms were significantly higher in chronic periodontitis, compared to both, healthy and gingivitis subjects (Ilgenli et al, 2006). Furthermore, chronic periodontitis subjects show higher MMP-13 activity in active sites compared to inactive sites and healthy controls.…”

“…MMP-13 has been implicated in bone resorptive process, along with MMP-9 (Hill et al 1995;Hill et al 1994;Holliday et al 1997). Total MMP-13 levels, as well as proenzyme (~60 kDa) and its active forms (~45-50 kDa), have been shown to increase in chronic periodontitis versus healthy sites in GCF in association with clinical periodontal parameters (Tervahartiala et al 2000;Kiili et al 2002;Ilgenli et al 2006). MMP-9 is the major gelatinase in oral fluids (Makela et al 1994).…”
