Gibberellin Response in Lettuce Hypocotyl Sections

Abstract: Excised lettuce (Lactuca sativa L.) hypocotyl sections retain the ability to elongate in response to gibberellic acid (GA3) addition. In 48 hr at 30 C a GA3-treated segment more than doubles while a control segment elongates less than 50%70. Auxin has no detectable effect on this system. Sensitivity to GA3 is not decreased by apex or root removal. Of the experimental variables tested, temperature, sucrose, and preincubation in water affect growth both with and without GA3. Blue and far red light inhibit growth… Show more

“…Potassium chloride has been shown to stimulate the growth of light-grown, GA-treated lettuce hypocotyl sections although KCl is without effect on the growth of sections incubated without GA (15). Our investigation shows that in addition to K+, Na+ and Rb+ cause significant growth promotion (Table I).…”

“…A stimulatory effect of 10 mm KCI on the elongation growth of excised lettuce hypocotyl sections was reported by Silk and Jones (15). Potassium chloride causes at least a 2-fold increase in the length of dark-grown sections and of GA-treated, lightgrown sections although it has no effect on the growth of lightgrown sections incubated in H20.…”

“…Cell wall plasticity of Avena internode segments, measured by instron extension and microextension of living internode segments, was increased 150% by treatment with GA, with a lag of about 1 h (1). ' Supported by a grant from Monsanto Co., St Excised hypocotyls oflettuce, incubated in the light, elongated in response to GA (21). The addition of 10 mM KCl doubled elongation for segments preincubated with GA but had no effect on the controls.…”

“…Thus GA3 affected both the mechanical (viscoelastic) and biochemical (chemorheological) properties of the cell walls of light-grown cucumber. The previous hypothesis, that GA3 stimulates cucumber hypocotyl growth by increasing osmotic pressure and cell turgor, is contradicted by our results.GA3 can reverse the inhibition of stem elongation caused by light in many plant species (1,12,17,18,21,23,24). The mechanism by which GA3 enhances the rate of elongation has been investigated by several groups, many of whom have concluded that GA3 acts on cell wall mechanical properties to increase growth (2,13,15,18,20).…”
