Although ghrelin acts as a modulator of feeding behavior and energy metabolism in the central nervous system, recent studies have implicated the peripheral actions of ghrelin in reproductive tissues. Here, we investigated the eff ects of chronic administration of ghrelin on the motility, plasma membrane integrity and concentration of rat spermatozoa. 45-d male Wistar rats were scheduled for the study and were divided into control and treatment groups. In the treatment group, 1 nmol of ghrelin was administered as sc injection for 10 consecutive days or vehicle (physiological saline) to the control rats. Sperm collection was achieved by killing of the rats on days 15, 25 and 50 after fi rst injection. Total sperm motility and forward progressive movement did not exhibit signifi cant diff erence during the experiment, although, there was a tendency for greater motion rate on d 15 and 25 in the treated rats compared to the control group. Plasma membrane integrity (HOS-reacted spermatozoa) was signifi cantly higher in the treated animals, especially on day 15 as well as day 25, because of possible antioxidant properties of ghrelin. Th is value was statistically higher on day 15 than that of day 25 (P <0.05). Likewise, there was a signifi cant correlation between the FPM (P <0.0001, r = 0.79) and TSM (P <0.01, r = 0.52) with the HOS test percentage in the treatment group. It was not observed statistically diff erence in the sperm concentration between groups during all of the experimental days. In conclusion, chronic administration of ghrelin (similar to induced by energy defi ciency such as fasting) increased the integrity of sperm membrane, however, the sperm motility and concentration did not display any alterations.

Ghrelin Antagonized 1-Methyl-4-Phenylpyridinium (MPP+)-Induced Apoptosis in MES23.5 Cells

Ghrelin is an endogenous ligand for the growth hormone secretagogue receptor (GHS-R) acting to stimulate growth hormone release. In the previous study, we have observed the neuroprotective effects of ghrelin on dopaminergic neurons in vivo in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine -treated Parkinson's disease mice. In order to illustrate the underlying mechanisms, in the present study, we conducted our experiment in vitro in 1-methyl-4-phenylpyridinium (MPP(+))-treated MES23.5 cells that could express GHS-R1a. Ten- to 1,000-micromol/L MPP(+) treatment caused decreased cell viability, with increased lactate dehydrogenase leakage. A 200-micromol/L MPP(+) treatment was chosen to do the further experiments. MES23.5 cells treated with 200 micromol/L MPP(+) showed decreased mitochondrial transmembrane potential, an elevated level of reactive oxidative species production and activation of caspase-3. Additionally, these cells also showed apoptotic morphological changes. Pretreatment with different doses of ghrelin (10(-12)-10(-7) mol/L) could abolish the MPP(+)-induced apoptotic changes in a dose-dependent manner. These results suggested that ghrelin could antagonize MPP(+)-induced apoptosis in MES23.5 cells. The protective effects of ghrelin involved the restoration of mitochondria function.

Ö ÖZ ZE ET T A Am ma aç ç: : Vitiligonun insülin direnci, lipid anormallikleri ve metabolik sendrom gibi metabolik bozukluklarla ilişkili olduğuna dair çeşitli çalışmalar vardır. Plazma grelin seviyeleri obez bireylerde düşük bulunmuş ve hiperinsülinemi ve insülin direnci gibi metabolik sendromun önemli bulgularından biri olarak kabul edilmiştir. Bu çalışmanın amacı, vitiligolu hastalarda serum grelin seviyelerini incelemektir. G Ge er re eç ç v ve e Y Yö ön nt te em ml le er r: : Çalışmaya 30 vitiligolu hasta ve 30 kontrol birey alındı. Açlık kan şekeri, serum lipidleri, insulin, C-peptid ve tiroid fonksiyon testlerine bakıldı. The current study aimed to investigate change in serum ghrelin levels in patients with vitiligo.
