Ghrelin against alendronate-induced gastric damage in rats

Abstract: Alendronate sodium, a primary amino bisphosphonate, is widely used in the treatment of various diseases that are associated with bone resorption, such as postmenopausal osteoporosis and Paget's disease of bone. Although the adverse effects of biphosphonates on the gastrointestinal system have been demonstrated in experimental and clinical studies, the exact mechanisms underlying this damage are not clear yet. Ghrelin, a 28 amino acid peptide produced predominantly by the stomach, was shown to exert a potent pr… Show more

“…However, the sperm motility and concentration did not exhibit signifi cant changes. Th ese results confi rm the recently reports concerning the antioxidative eff ects of ghrelin in the other tissues (Zwirska-Korczala et al, 2007 ;Kawczynska-Drozdz et al, 2006 ;Iseri et al, 2005 ).…”

“…Mitochondria is the major site of ROS production and also prime target of oxidative molecular damage (Chan 2004;Yao et al 2006), the consequent formation of ROS further damages the mitochondrial membrane and such damages are implicated as key events in the pathogenic cascades leading to apoptosis. Ghrelin is known to inhibit H 2 O 2 -induced cytokine release in human endothelial cells (Li et al 2004) and to ameliorate oxidative gastric damage induced by alendronate (Iseri et al 2005). These results suggest that ghrelin may have antioxidant properties.…”
