Germline Neurofibromin 1 mutation enhances the anti‐tumour immune response and decreases juvenile myelomonocytic leukaemia tumourigenicity

Abstract: Summary Juvenile myelomonocytic leukaemia (JMML) is an aggressive paediatric leukaemia characterized by mutations in five canonical RAS pathway genes, including the NF1 gene. JMML is driven by germline NF1 gene mutations, with additional somatic aberrations resulting in the NF1 biallelic inactivation, leading to disease progression. Germline mutations in the NF1 gene alone primarily cause benign neurofibromatosis type 1 (NF1) tumours rather than malignant JMML, yet the underlying mechanism remains unclear. Her… Show more

“…As a new way to treat tumors with immunotherapy, mannose-modified lipid calcium phosphate nanoparticle vaccine has shown great promise in improving the immune response to tumors and building immune memory [231][232][233][234][235]. This provides new ideas and strategies for the future treatment of cancer and is expected to play an important role in clinical practice, bringing more effective treatment effects and a better quality of life for patients [236][237][238][239][240].…”

“…In contrast, germline mutations are inherited changes that affect reproductive cells and are present in all somatic cells; they may be common or rare in a given population. Of note, not only are somatic variants important for cancer treatment and mainly used as molecular targets, but germline mutations may also contribute, at least in part, to tumor development, progression, and resistance ( Chen X et al, 2023 ; Wang et al, 2023 ).…”
