Genotypic to Expression Profiling of Bovine Calcium Channel, Voltage-Dependent, Alpha-2/Delta Subunit 1 Gene, and Their Association with Bovine Mastitis Among Frieswal (HFX Sahiwal) Crossbred Cattle of Indian Origin

Abstract: Calcium channel, voltage-dependent, alpha-2/delta subunit 1 (CACNA2D1) gene is considered to be an important noncytokine candidate gene influencing mastitis. Scanty of reports are available until today regarding the role play of CACNA2D1 gene on the susceptibility of bovine mastitis. We interrogated the CACNA2D1 G519663A [A>G] SNP by PCR-RFLP among two hundreds Frieswal (HF X Sahiwal) crossbred cattle of Indian origin. Genotypic frequency of AA (51.5, n=101) was comparatively higher than AG (35, n=70) and GG (… Show more

“…Our examination of the cellular calcium flux suggests that MA, but not LA, recognizes FFAR1/GPR40 in BUVECs. The CACNA2D1 gene encodes the alpha 2/delta subunit which is an accessory part of the voltage-gated calcium channel family (e.g., LTCC) and considered to be an important candidate protein influencing bovine mastitis [ 35 ]. Although many publications have reported LTCC activity in bovine immune cells, there is not yet any evidence that these channels are expressed in bovine endothelial cells.…”

“…Deb et al . [ 45 ] from their study of SNP detection by RT-PCR in bovine CACNA2D1 gene (chromosome 4) at G519663A locus, demonstrated that GG genotype is associated with the lowest somatic cell score (SCS), and the transcript abundance of CACNA2D1 mRNA and was favorable for the mastitis resistance. Similar finding with the help of PCR-RFLP was observed by Yuan et al .…”

“…Calcium channel, voltage-dependent, alpha-2/delta subunit 1 residing on chromosome 4, containing 42 exons, is another key genetic signature with an important role in clinical mastitis. It has been documented that SNP-G519663A at exon 18 (80), SNP-G519663A, and SNP-A526745G at exon 24 in the bovine CACNA2D1 gene were significantly linked with lower milk SCS levels and mastitis resistance in dairy cattle (82, 102). Moreover, it has also been noticed that SNP G38819398A at exon 18 (81), variant-T38826986G at exon 19 (103), and SNP-C367284A of CACNA2D1 (104) were positively linked with milk SCS in Sahiwal and Karan Fries.…”
