Trametes hirsuta and a purified laccase from this organism were able to degrade triarylmethane, indigoid, azo, and anthraquinonic dyes. Initial decolorization velocities depended on the substituents on the phenolic rings of the dyes. Immobilization of the T. hirsuta laccase on alumina enhanced the thermal stabilities of the enzyme and its tolerance against some enzyme inhibitors, such as halides, copper chelators, and dyeing additives. The laccase lost 50% of its activity at 50 mM NaCl while the 50% inhibitory concentration (IC 50 ) of the immobilized enzyme was 85 mM. Treatment of dyes with the immobilized laccase reduced their toxicities (based on the oxygen consumption rate of Pseudomonas putida) by up to 80% (anthraquinonic dyes). Textile effluents decolorized with T. hirsuta or the laccase were used for dyeing. Metabolites and/or enzyme protein strongly interacted with the dyeing process indicated by lower staining levels (K/S) values than obtained with a blank using water. However, when the effluents were decolorized with immobilized laccase, they could be used for dyeing and acceptable color differences (⌬E*) below 1.1 were measured for most dyes.It is known that 90% of reactive textile dyes entering activated sludge sewage treatment plants will pass through unchanged and will be discharged to rivers (34). Not all dyes currently used could be degraded and/or removed with physical and chemical processes, and sometimes the degradation products are more toxic (40). The traditional textile finishing industry consumes about 100 liters of water to process about 1 kg of textile materials. New closed-loop technologies such as the reuse of microbially or enzymatically treated dyeing effluents could help to reduce this enormous water consumption.Several combined anaerobic and aerobic microbial treatments have been suggested to enhance the degradation of textile dyes (5, 23, 32). However, under anaerobic conditions, azo-reductases usually cleave azo dyes into the corresponding amines, many of which are mutagenic and/or carcinogenic (10,11,32). Furthermore, azo reductases have been shown to be very specific enzymes, thus cleaving only azo bonds of selected dyes (50, 51). By contrast, laccases act oxidatively and less specifically on aromatic rings, thus having potential to degrade a wider range of compounds (43).Laccases are involved in the biodegradation of lignins, which constitute the main noncarbohydrate component in wood and are among the most abundant groups of biopolymers in the biosphere. Two known mutagenic drugs, cyclophosphamide and vinblastine sulfate, were tested using the micronucleus test in the native fish species, Astyanax bimaculatus, in order to determine which of these drugs and the doses which would be the most adequate for use as positive controls in this species. This Brazilian fish species was chosen because few toxicity studies have used native fish species and this particular species is widely consumed in various regions of Brazil. Three thousand erythrocytes per specimen were scored. Doses of 16 and 8 mg/kg body weight of cyclophosphamide and vinblastine sulfate, respectively, were the most effective in causing micronuclei. Cyclophosphamide was the most mutagenic of the two drugs and is recommended for use as a positive control in A. bimaculatus.
Duas drogas reconhecidas como mutagênicas, ciclofosfamida e vimblastina sulfato, foram avaliadas usando o teste do micronúcleo em uma espécie de peixe nativa, Astyanax bimaculatus, para detectar que droga e quais doses são as mais adequadas para serem usadas como controles positivos para esta espécie. Esta espécie de peixe brasileira foi escolhida devido à escassez de estudos toxicológicos com espécies de peixes nativos e também porque ela é amplamente consumida em algumas regiões do Brasil. Um total de 3000 eritrócitos por espécimen foram contados. As doses de 16 e 8 mg/kg de peso corporal de ciclofosfamida e de vimblastina sulfato, respectivamente, foram as mais efetivas na indução de micronúcleos. A ciclofosfamida mostrou ser o melhor agente mutagênico para ser usado como um controle positivo para Astyanax bimaculatus

RÉSUMÉObjectif : Les effets cytogénotoxiques des sites du PONT et d'ACCRON de la lagune de Porto-Novo à des concentrations de 0%, 25%, 50%, 75%, 100% ont été évalués par le test de toxicité basé sur les cellules des racines des oignons. Méthodologie et résultats : Dans cette étude, la longueur des racines et le test sur les aberrations chromosomiques ont été utilisés pour déterminer la concentration efficace (96h, CE50), l'inhibition de la croissance des racines, l'indice mitotique et le taux d'aberration chromosomique. D'après les résultats obtenus, les eaux du site du PONT sont 2,24 fois plus toxiques que celles des eaux du site d'ACCRON et on a une différente significative entre la moyenne des longueurs des racines des oignons exposés dans les différentes concentrations des deux sites. Cela indique que l'inhibition de la croissance des racines dépend des concentrations. L'indice mitotique (IM) croit avec l'augmentation des concentrations des eaux sur le site d'ACCRON par contre sur le site du PONT, il diminue de 0% à 25% et croît de 50% à 100%. Les eaux des deux sites ont provoqué des aberrations chromosomiques des cellules des racines des oignons avec des malformations chromosomiques telles que les chromosomes en fragments de pont, vagabonds et collants. Les aberrations communes observées lorsque les concentrations sont faibles, sont les chromosomes en fragments de pont, visqueux, vagabonds et les anaphases polaires. Le test de génotoxicité effectué sur les chromosomes des racines des oignons a permis de mesurer les effets génotoxiques des eaux des deux sites étudiés. Conclusion et application des résultats : Les tests de cytogénotoxicité basés sur les racines d'oignons effectués sur les eaux des deux sites ont montré d'une part que les deux sites sont génotoxiques et que le site du PONT est plus toxique que celui d'ACCRON d'autre part.
