Genotoxic effects of three selected black toner powders and their dimethyl sulfoxide extracts in cultured human epithelial A549 lung cells in vitro

Abstract: Until now, the adverse effects of toner powders on humans have been considered to be minimal. However, several recent reports have suggested possible significant adverse health effects from toner dust inhalation. The aim of this study was to evaluate the genotoxic potential of black toner powders in vitro. For the study of DNA damage, A549 cells were exposed to toner-powder suspensions and to their DMSO extracts, and then subjected to the comet assay and to the in-vitro cytokinesis block micronucleus test (CB-… Show more

“…within airborne PM, pollution-derived magnetite nanoparticles might require consideration as a possible AD risk factor. In addition to occupational settings [including, for example, exposure to printer toner powders (36)], higher concentrations of magnetite pollution nanoparticles are likely to arise in the indoor environment from open fires (25) or poorly sealed stoves used for cooking and/or heating, and in the outdoor environment from vehicle (especially diesel) and/or industrial PM sources.…”

“…Subsequently, they were kept under constant light in sterile falcon tubes (50ml). From day zero cells were counted with the FlowCam using the vitality dye Erythrosine B (4 mg ml -1 in 1xPBS) following the method of 61 . This staining is based on the differential uptake ratio of erythrosine between dead/alive cells.…”
