Genomic organization of mouse Fc gamma receptor genes.

Abstract: We have isolated and characterized the gene coding for the mouse Fc receptor that is termed Fc,,RHa. The gene contains five exons and spans approximately 9 kilobases.

“…Analysis of their genomic structure further supports the relationship not only between the FcRH genes, but with their FcR neighbors as well. A conserved feature among the Fc␥RI, Fc␥RII, Fc␥RIII, and FcR genes is a signal peptide encoded by two separate exons, the second of which (S2) is a 21-bp ''miniexon'' (43)(44)(45). The conservation of the 21-bp S2 miniexon for all FcRH and FcR family members on chromosome 1 contrasts with the 36-bp S2 exon encoding the second half of the signal peptide for the Fc␣R gene on chromosome 19q13 (46).…”

“…The first exon, 5Ј UT͞S1, encodes the 5Ј UT region, the ATG translation initiation codon, and the first half of a split signal peptide. S2, the second exon, is separated from 5Ј UT͞S1 by a long intron of Ϸ12.9 kb and, like the neighboring FcRs, is 21 bp in length (43)(44)(45). The extracellular region is encoded by three closely clustered exons, EC1-EC3, that code for the three Ig-like domains.…”

“…In contrast to wild-type C57BL/6 mice, however, C57BL/6 mice homozygous for Fc + RIII § chain -/selectively express the 129-derived Ly-17.1 alleles of Fc + RII reflecting a tight genetic linkage of Fc + RII and Fc + RIII. In accordance, Fc + RII and Fc + RIII genes had been described to be very closely linked with a maximal distance of 160 kb on mouse chromosome 1 [37]. All mice were bred and maintained under dry barrier conditions in the animal facilities at the Hannover Medical School (Hannover, Germany).…”

“…It has been suggested that the formation of this cluster has included at least one evolutionarily very recent duplication event, which has resulted in very high sequence identity (197%) over large portions of the most recently duplicated genes and their surrounding non-coding sequence [39]. In addition, while humans have five low-affinity IgG receptor genes (FCGR2A, FCGR2B, FCGR2C, FCGR3A and FCGR3B), the mouse has only three (fcgr2a, fcgr2b, fcgr3) [38,52]. Our analysis of the available human sequence suggests that this region is organized into two approximately 81-kb blocks, with high sequence identity.…”
