Genomic instability and innate immune responses to self-DNA in progeria

Abstract: In the last decade, we have seen increasing evidence of the importance of structural nuclear proteins such as lamins in nuclear architecture and compartmentalization of genome function and in the maintenance of mechanical stability and genome integrity. With over 400 mutations identified in the LMNA gene (encoding for A-type lamins) associated with more than ten distinct degenerative disorders, the role of lamins as genome caretakers and the contribution of lamins dysfunction to disease are unarguable. However… Show more

“…Tumor immunology is a trending topic in cancer research. Immune-related mechanisms play important roles in multiple pathophysiological progresses (Rožman, 2018;Gonzalo and Coll-Bonfill, 2019;Walker et al, 2019), and immunotherapeutic strategies are considered as a promising direction for the treatment of many cancers (Washah et al, 2020). Immunotherapy, such as programmed death-1 (PD-1) and programmed death ligand-1 (PD-L1) inhibitors, plays a good anti-tumor effect in the treatment of malignant tumors, such as lung cancer (Cheng et al, 2020;Eguren-Santamaria et al, 2020) and melanoma (Cuevas and Daud, 2018;Albershardt et al, 2020).…”

“…Janus kinases (JAKs) are major activators of signal transducers and play a significant role in cellular biological processes, inflammation, and immunity [ 4 7 ]. JAK/STAT signaling is a key regulator of gene expression, transcriptional programs, and immune response.…”

“…Thus, a signature based on multi-gene cooperation can improve the predictive ability of each gene, resulting in a more reliable and robust predictive ability versus a single biomarker (Ashley 2015;Jiang et al 2020;Wolfe et al 2020;Zhu et al 2016). DNA damage repair genes are needed to maintain cell homeostasis and are extensively involved in cell replication, chromatin modification, DNA repair, checkpoint signaling, reactive oxygen species, metabolism, senescence, and apoptosis cellular mechanisms (Brinkman et al 2020;Csiszar et al 2019;Gonzalo & Coll-Bonfill 2019;Lee et al 2018;Royce et al 2019;Ungvari et al 2019). Genetic mutations can effectively induce tumorigenesis and progression (Pearl et al 2015).…”
