Genomic Characterizations of Six Pigeon Paramyxovirus Type 1 Viruses Isolated from Live Bird Markets in China during 2011 to 2013

Abstract: The genomes of six pigeon paramyxovirus type 1 (PPMV-1) isolated from symptomless pigeons in live poultry markets during the national active surveillance from 2011 to 2013 were sequenced and analyzed in this study. The complete genome lengths of all isolates were 15,192 nucleotides with the gene order of 3’-NP-P-M-F-HN-L-5’. All isolates had the same motif of 112RRQKRF117 at the cleavage site of the fusion protein, which was typical of velogenic Newcastle disease virus (NDV). Several mutations were identified … Show more

“…The cross-reactivity HI assay showed that the R value between the GZ08 strain and the commonly used vaccine LaSota was 0.59, indicating a minor antigenic difference between LaSota and GZ08. The results were similar to those of previous research [24,34]. A large number of reports confirmed that antigen matching between vaccine and epidemic strains could increase the effectiveness of vaccines against epidemic strains [51][52][53][54].…”

“…The cross-reactivity HI assay between the commonly used vaccine LaSota and the PPMV-1 strain, GZ08, was conducted as described previously [34]. Briefly, 3-week-old pigeons were intramuscularly injected with inactivated oil-emulsified LaSota or GZ08 virus.…”

“…Since the neutralization antibodies raised against the F protein define the protection status of the host, these modifications in the F protein explain the potential of immune escape and lack of sterile immunity caused by currently available vaccines. Moreover, the substitutions particularly in the fusion peptide, hydrophobic regions and transmembrane region of the F protein and neutralizing epitopes of the HN protein could result in altered fusion activity and neutralizing escape variants [14,15,37]. Taken together, nucleotide and subsequent amino acid substitutions explained the evolving nature of RNA viruses including the AAvV-1 however warrant future investigation to delineate the importance of these mutations in the pathobiology of AAvV-1 [16,20,36].…”

“…The egg lethal dose 50 (ELD50) was calculated by Reed and Muench method [29]. Hemagglutination inhibition (HI) assay was performed as previously described [26,37] using four hemagglutinating units for LaSota, NDV/Cattle egret/Egypt/SDU-1/2016 and NDV/Teal/Egypt/SDU-4/2016 and antisera collected from SPF chickens vaccinated with LaSota (Genotype II) live attenuated and commercial killed vaccine (Genotype VII).…”

“…PPMV-1 was introduced to Hong Kong in 1985, subsequently spread throughout China, and now poses a serious threat to the Chinese pigeon breeding industry and wild birds population [12]. The virus is now one of the most destructive pathogens for pigeons in the country, also affecting pigeon farms with increased frequency (Awu et al 2015; Guo et al 2013;Ren et al 2016;Wei et al 2018).…”
