Genomic characteristics and comparative genomics analysis of Penicillium chrysogenum KF-25

Abstract: BackgroundPenicillium chrysogenum has been used in producing penicillin and derived β-lactam antibiotics for many years. Although the genome of the mutant strain P. chrysogenum Wisconsin 54-1255 has already been sequenced, the versatility and genetic diversity of this species still needs to be intensively studied. In this study, the genome of the wild-type P. chrysogenum strain KF-25, which has high activity against Ustilaginoidea virens, was sequenced and characterized.ResultsThe genome of KF-25 was about 29.… Show more

“…For instance, a comparison between two strains of Penicillium chrysogenum from China and United States revealed a very low level of sequence divergence between the two strains measured as the density of single nucleotide polymorphisms (SNP) between the two strains (∼0.08 SNPs/kb; Wang et al 2014 ). In contrast, a comparison of the P. chrysogenum type strain to P. chrysogenum KF-25 showed a high level of protein sequence divergence where proteins shared only 75.1% similarity on average, suggesting they are actually different species ( Peng et al 2014 ). In Aspergillus fumigatus , the comparison of two strains, A1163, a derivative strain of CBS144-89/CEA10 isolated from a patient in Paris, and Af293 isolated from a patient in United Kingdom, showed an identity of 99.8% at nucleotide level between the core genes ( Fedorova et al 2008 ).…”

“…Soil fungi produce a wide range of chemical compounds that have been utilized by humans for many years, such as monascorubrin and its derivatives from Monascus species, which serve as natural red colorants for food (5), and penicillin and derived ␤-lactam antibiotics produced by Penicillium chrysogenum (6). Soil fungi can also produce mycotoxins that contaminate human and animal food and that represent a threat to human and animal health (7).…”

“…It is also known that in all these strains, P. chrysogenum Wis 54-1255, A. chrysogenum RNCM 408D and A. terreus No. 44-62, the only one copy of corresponding BGC is present, one copy of pen genes [86], one copy of "early" and "late" cef genes [32,41] and one copy on lov genes [84] respectively. In this regard, the CSI programs for these strains could follow a rather different pathway than P. chrysogenum DS17690 with eight copies of pen genes [30], without significant shifting and removing the global regulation of SM and mutation in LaeA.…”
