Genome-wide analyses of long noncoding RNA expression profiles in lung adenocarcinoma

Abstract: LncRNAs have emerged as a novel class of critical regulators of cancer. We aimed to construct a landscape of lncRNAs and their potential target genes in lung adenocarcinoma. Genome-wide expression of lncRNAs and mRNAs was determined using microarray. qRT-PCR was performed to validate the expression of the selected lncRNAs in a cohort of 42 tumor tissues and adjacent normal tissues. R and Bioconductor were used for data analysis. A total of 3045 lncRNAs were differentially expressed between the paired tumor and… Show more

“…28 Peng et al analyzed aberrant lncRNA genes in LAD and found that the top GO terms are mainly enriched in cell adhesion, parts of the extracellular region, and cytoskeletal protein binding. 29 In our comparison of LAD with and without lymphatic metastasis, we found that cell-cell adhesion was downregulated in BP of GO, and when altered may facilitate the migration and metastasis of tumor cells. Upregulated genes in this comparison were enriched in mostly intracellular rather than extracellular regions where the downregulated genes were found.…”

“…LncNONHSAT081507.1 (LINC81507) was first identified by Peng et al using Agilent Human LncRNA Array. 44 Recently, Peng et al found reduced expression of LINC81507 resulted in cell growth, proliferation, migration and EMT in NSCLC cells, whereas ectopic overexpression of LINC81507 resulted in the opposite effects both in vitro and in vivo. 45 LINC81507 acted as a ceRNA for miR-199b-5p through directly binding and interfering miR-199b-5p-mediated regulation of CAV1 to reduce migration and invasion.…”

“…8 Accumulative evidences demonstrated that lncRNA could also act as a sponge for microRNA (miRNA) to suppress the target gene of the miRNA, which is considered to be a competing endogenous RNA (ceRNA). [10][11][12][13] Among the lncRNAs, colon cancer-associated transcript-1 (CCAT1) was proved highly associated with human cancers, 14 including gastric cancer, 15 ovarian cancer, 16 breast cancer, 17 and lung cancer. “…2 Recently, more and more studies indicated that the aberrations of lncRNA expression were found in many types of cancers involving in regulating the cell proliferation and metastasis, which will lead normal cells or tissues to tumorigenesis. [10][11][12][13] Among the lncRNAs, colon cancer-associated transcript-1 (CCAT1) was proved highly associated with human cancers, 14 including gastric cancer, 15 ovarian cancer, 16 breast cancer, 17 and lung cancer. 18 Specifically, lncRNA-CCAT1 has been reported to promote tumorigenesis and development in colorectal cancer.…”
