Genome Size and Karyotype Studies in Five Species of Lantana (Verbenaceae)

Abstract: Lantana species are an important component of the U.S. environmental horticulture industry. The most commonly produced and used species are L. camara and, on a smaller scale, L. montevidensis. Both were introduced to the United States from Central and/or South America. Lantana species native to the continental United States include L. canescens, L. depressa, L. involucrata, etc. and most of them have not been well exploited. This study was conducted to obtain information about somatic chromosome numbers, karyo… Show more

“…and tomato internal standard were finely cut and directly added to a petri dish containing one ml of LB01 buffer.. The homogenate was filtered through a nylon mesh (50 µm) and 50 µl of the DNA fluorochrome, propidium iodide (Sigma-Aldrich, St. Louis, MO, U.S.; 1 mg•ml −1 ) and RNase (Sigma-Aldrich, St. Louis, MO, U.S.; 1 mg•ml −1 ) were added to the filtered homogenate 54 . Three biological replicates of the two unreported species (S. arenicola and S. integrifolia) and a plant each for S. baicalensis and S. lateriflora were analyzed.…”

“…Study of the karyotype features of Lonicera followed the root tip squashing method for sampling, fixation, dissociation, staining, measuring and photographing (Parrish et al 2021; Wu et al 2021). When the root length of Lonicera seeds was up to 0.5–1.0 cm, 20 root tips were taken and pressed into slices.…”

“…Chromosome karyotype characteristics of Lonicera L.: Refer to the root tip squashing method for sampling, fixation, dissociation, staining, measuring and photographing (Parrish et al 2021;Wu et al 2021). When the root length of Lonicera L. seeds was up to 0.5-1.0 cm, 20 root tips were taken and pressed into slices; the numbers of clear and scattered chromosomes in the cells was counted when more than 85% of the cells have a constant and consistent chromosome numbers in 20 cells in each species Lonicera L., then the photos were taken under the 100 times oil lens (Ferreira et al 2020;Ruiyang Chen 2003;Yonggang Qiao et al 2020).…”
