Genome sequence and characterization of Streptomyces phage Pablito, representing a new species within the genus Janusvirus

Abstract: Streptomycetes are ubiquitous soil bacteria. Here we report the complete and annotated genome sequence and characterization of Streptomyces phage Pablito, isolated from a soil sample in Haarlem, the Netherlands using Streptomyces lividans as host. This phage was able to infect a diverse range of Streptomyces strains, but none belonging to the sister genus Kitasatospora. Phage Pablito has double-stranded DNA with a genome length of 49,581 base pairs encoding 76 putative proteins, of which 26 could be predicted.… Show more

“…Since the full nucleotide sequence was not similar to any phages, we performed phylogenetic analysis on the major capsid protein (GenBank: WAX22437.1), which is identified in most phage genomes and has been used previously for phylogenetic analysis 20 , 21 . There were 62 additional phage proteins with high percent amino acid identity to the major capsid protein encoded by MiCath.…”

“…The one-step growth curve of Vanseggelen and Verabelle showed a long latent period of 140 minutes and a relatively low burst size, even when compared to other Streptomyces phages. 5,21 The low burst size could be the result of the morphological complexity of Streptomyces 21 . The meaning of MOI loses its signi cance once spores have germinated and a mycelium is formed as multiple phages are able to attach, but the mycelium would still be counted as one colony forming unit 13,22 .…”

“…Streptomyces phages Vanseggelen and Verabelle were isolated from a soil sample obtained from the National Park Zuid-Kennemerland in the Netherlands (N52°23′31″, E4°34′49″) using Streptomyces coelicolor as the host. Details on isolation procedures are reported previously in detail 5 . The collected phage stock solutions were stored at 4°C.…”

“…The last few years have been marked by a sustained effort in the isolation and sequencing of Streptomyces phages from environmental samples (1719, 10, 20, 21). Moreover, the Streptomyces genome can constitute per se a viral genetic reservoir, through the genomes of temperate prophages that they may host.…”
