The pathogenic nature ofMorganella morganiito fruit flies has been described earlier; its biofilm prevents the larvae of mass reared fruit flies from proceeding to their next life stage by trapping them in their polymeric matrix. For the Mediterranean (Ceratitis capitata) fruit fly (“medfly”) mass rearing plants, this can be problem, due to losses generated in the production process. The presence of this bacteria is prevented by using different disinfectants, by themselves, or combined, which can act as selective pressure for the biofilm to resist the disinfectants. In this paper, we propose a method to isolate lytic bacteriophages and use them as quorum quenchers, to prevent microbial consortium from forming biofilms. When tried as viral cocktails, a very significant reduction of biofilm was found. However, when used with one of the two tried industrial disinfectants, the viral effect wasn’t as significant. When interpreted alongside the first experiments, this result opens the door to use bacteriophages to reduce the minimum inhibitory concentration necessary of the disinfectant. A reduction of said concentration would translate into less selective pressure for a resistance against it, and a reduction in costs for the mass rearing process at the time of implementation of the system.

Lactobacilli are ubiquitous in nature and symbiotically provide health benefits for countless organisms including humans, animals and plants. They are vital for the fermented food industry and are being extensively explored for healthcare applications. For all these reasons, there is considerable interest in enhancing and controlling their capabilities through the engineering of genetic modules and circuits. One of the most robust and reliable microbial chassis for these synthetic biology applications is the widely used Lactiplantibacillus plantarum species. However, the genetic toolkit needed to advance its applicability remains poorly equipped. This mini‐review highlights the genetic parts that have been discovered to achieve food‐grade recombinant protein production and speculates on lessons learned from these studies for L. plantarum engineering. Furthermore, strategies to identify, create and optimize genetic parts for real‐time regulation of gene expression and enhancement of biosafety are also suggested.

Bu çalışmada Batı Karadeniz Bölgesi’nde, beş ilden toplanan, geleneksel yöntemlerle üretilmiş yoğurtlardan laktik asit bakterilerinin izolasyonu, tanımlanması, bazı starter kültür özelliklerinin belirlenmesi amaçlanmıştır. Suşların tanımlanması MALDI-TOF MS Biotyper sistemi ile gerçekleştirilmiştir. İzole edilen 84 izolattan 2’si Lactobacillus helveticus, 2’si Lactiplantibacillus plantarum, 3’ü Limosilactobacillus fermentum, 1’i Enterococcus faecalis ve 76’sı Lactobacillus delbrueckii olarak tanımlanmıştır. İzolatların eritromisin, teikoplanin, streptomisin, rifampisin, amfisilin, klindamisin, sefotaksim, kloramfenikol, tetrasiklin ve vankomisine karşı duyarlı, nalidiksik asit, siprofloksasin, ofloksasin, gentamisin ve trimethoprim sülfametoksazole karşı dirençli oldukları tespit edilmiştir. İzolatların Salmonella Typhimurium ve E. coli O157:H7’ye karşı antimikrobiyal etki gösterdiği ancak Bacillus cereus, Listeria monocytogenes ve Staphylococcus aureus’a karşı antimikrobiyal etkisinin düşük olduğu tespit edilmiştir. İzolatlardan 8’inin ürettiği asitliğin %0.9 ve 0.95 arasında olduğu bulunmuştur. Ayrıca 74 izolatın diasetil üretimi gerçekleştirdiği belirlenmiştir. Starter olma potansiyeli yüksek 26 suşun proteinaz aktivitesi ve fajlara karşı direnç durumu incelenmiştir. Suşlardan 5’inin proteolitik aktivite göstermediği ve faj direnci testi sonuçlarına göre izolatlardan 8 tanesi Ф709-X1 fajına karşı duyarlı olup, 12 adet Lactobacillus delbrueckii izolatının ise çalışmada kullanılan 15 farklı bakteriyofaja karşı dirençli oldukları belirlenmiştir. Çalışma sonuçlarına göre izole edilen kültürlerin incelenen özellikler açısından starter kültür olarak kullanılabilme potansiyeline sahip oldukları belirlenmiştir.
