Genome Properties of the Diatom Phaeodactylum tricornutum

Abstract: Diatoms are a ubiquitous class of microalgae of extreme importance for global primary productivity and for the biogeochemical cycling of minerals such as silica. However, very little is known about diatom cell biology or about their genome structure. For diatom researchers to take advantage of genomics and post-genomics technologies, it is necessary to establish a model diatom species. Phaeodactylum tricornutum is an obvious candidate because of its ease of culture and because it can be genetically transformed… Show more

“…Swain, 1999;Kavanagh et al, 2001;Stein et al, 2003;Finlay et al, 2005;Sun et al, 2005). The availability of a good model system in P. tricornutum, with publication of its complete genome sequence, and the development of microarrays (Scala et al, 2002;Montsant et al, 2005), now facilitates the analysis of surface preferences for adhesion through molecular genetic and gene expression studies. Microarrays can be used in 'expression analysis' or expression profiling.…”

“…Diatoms responsible for biofouling not only differ in adhesive strength depending on the surface they are exposed to but also in terms of the difference in adhesive strength between species (Holland et al, 2004). The isolate Pt 1.8.6 has been used in nearly all of the P. tricornutum studies described in the literature (Borowitzka & Volcani, 1978;Scala et al, 2002;Montsant et al, 2005;Dugdale et al, 2006). It is from this isolate that the genome has been sequenced and the EST libraries created (Montsant et al, 2005).…”

“…Previous work by Grö ne and found DMSP synthesis to be inhibited by addition of protease inhibitors, indicating that protein turnover was a source for DMSP precursors. FCPs are the most abundant transcripts in EST libraries (Scala et al, 2002;Mock et al, 2005). Furthermore, incubations with H 14 CO 3 2 followed by autoradiography of membrane proteins separated by denaturing PAGE found protein bands corresponding to the 17.5-and 18.5-kD bands detected by FCP antibody account for 6% of membrane protein synthesis under high-light versus 18% under low-light conditions (Friedman and Alberte, 1986).…”
