Genome analysis of Excretory/Secretory proteins in Taenia solium reveals their Abundance of Antigenic Regions (AAR)

Abstract: Excretory/Secretory (ES) proteins play an important role in the host-parasite interactions. Experimental identification of ES proteins is time-consuming and expensive. Alternative bioinformatics approaches are cost-effective and can be used to prioritize the experimental analysis of therapeutic targets for parasitic diseases. Here we predicted and functionally annotated the ES proteins in T. solium genome using an integration of bioinformatics tools. Additionally, we developed a novel measurement to evaluate t… Show more

“…The AAR value for the nTsKE7 protein was 44.2, as estimated by BepiPred analysis. This value is lower than the previously established AAR value for the antigenic experimental T. solium secretome, 81.3 (24), which suggests a higher density of antigenic regions in the nTsKE7 protein. This result is consistent with the immunogenic properties reported herein for the rTsKE7 protein.…”

“…The threshold score was set to 0.35, and only those antigenic regions with 6 or more amino acids in length were selected. Then, an abundance of antigenic regions (AAR) value was calculated to normalize the number of antigenic regions according to the protein length, as previously reported (24).…”

“…We wanted to get the maximum score for each epitope so as to get the most stable structure of mRNA that would lead to the most antigenic epitope for building our immunodiagnostic platform for assessing stress in poultry. Other studies have shown a high performance using this analysis and the determination of the best antigenic epitope relevant for an immunodiagnostic platform, vaccine production, and several treatments, especially in humans (Kolaskar and Tongaonkar, 1990;Hernández-Guzmán et al, 2014;Cai et al, 2015;Gomez et al, 2015). Such results could start the process for understanding of the immunomodulation caused by stress.…”

“…The more antigenic a protein the more likely it can be used as a biomarker, targeted for immunotherapy, or used in vaccinations. To help prioritize Aspergillus proteins, the Abundance of Antigenic Regions (AAR) 24 value of proteins was calculated using two methods, Kolaskar-Tongaonkar 90 and BepiPred 2.0 91 (Methods). The lower the AAR value the more antigenic the protein 24 .…”
