Genome analysis of a G9P[23] group A rotavirus isolated from a dog with diarrhea in China

Abstract: Genotype G9 is an emerging genotype among species A rotavirus (RVA) circulating in humans and pigs worldwide. In this study, an RVA strain designated RVA/Dog-tc/CHN/SCCD-A/2017/G9P[23] was isolated in cell culture from a pet dog stool sample with acute diarrhea, and its whole genome was sequenced. The genotype constellation of SCCD-A was G9-P[23]-I5-R1-C1-M1-A8-N1-T1-E1-H1. All genome segments except the VP1 gene were closely related to the genes from porcine RVA strains or porcine-like human RVA strains. On t… Show more

“…The evolutionary analysis revealed that rotavirus G1 and G3 strains were dominant before 2017, while after 2018 there was a surge of human G9 strain. The G9 strain, meanwhile, has been present and dominant in swine and dogs [ 51 , 54 ]. Previous studies have revealed that the G3 and G9 strains of RAV cause diarrhea in human infants and young children, and the recombination of RVA in humans and animals causes the generation of new PoRVA genotypes [ 55 , 56 , 57 , 58 , 59 ].…”

“…In contrast, sequence variation among strains between lineages was considerably higher, ranging from 5% to 14% (Phan et al, 2007). The number of a novel sub‐lineage of G9 lineage III has increased continuously in recent years, and has become phylogenetically more complex (Yan et al, 2019). In this study, the three G9 RVAs were assigned to different lineages, and belonged to lineages III and VI.…”

“…The cDNA was stored at −20°C. To determine the viral genome sequences in the RVA‐positive samples, the complete sequences of the three porcine RVAs were amplified with RT–PCR using 27 pairs of primers (Yan et al, 2019). The PCR products were purified, cloned into the pMD19‐T vector (TaKaRa) and transformed into Escherichia coli competent cells DH5α for Sanger sequencing.…”

“…The second example is that through the phylogenetic tree analysis of the genotypes of HRV in Wuhan from 2000 to 2013 [48], it was found that the G3P [8] type HRV was a RV from other species. The third example is the first successful isolation of the G9P [23] CRV strain from dog faeces by Yan and colleagues; the G9 genotype is widely prevalent in humans and pigs [2].…”

“…Rotavirus is the genus of the family Reoviridae with viruses that are a major cause of gastroenteritis in infants and young animals worldwide [1][2][3]. Rotavirus causes diarrhoea in many animals [4][5][6][7][8][9].…”
