Genistein decreases the breast cancer stem-like cell population through Hedgehog pathway

Abstract: IntroductionThe existence of breast cancer stem-like cells (BCSCs) has profound implications for cancer prevention. Genistein, a predominant isoflavone found in soy products, has multiple robust anti-tumor effects in various cancers, especially in the breast and prostate cancer. In this study, we aimed to evaluate genistein inhibition of BCSCs and its potential mechanism by culturing MCF-7 breast cancer cells and implanting these cells into nude mice.MethodsCell counting, colony formation and cell apoptosis an… Show more

“…Thus, genistein, a predominant isoflavone found in soy products, has been reported to decrease the CD44/CD24-positive CSC population in MCF7 breast tumor cells and mammosphere formation by downregulating Hedgehog-Gli1 signaling pathway (Fan et al, 2013). Acting on the same signaling pathway in prostate cancer, genistein was shown to suppress prostatosphere and colony formation in vitro and tumorigenicity in vivo (Zhang et al, 2012a).…”

“…8 While genistein has been reported to have a radioprotective effect on normal tissues, 9 treatment of non-inflammatory breast cancer cells with high doses (µM range) of genistein has been shown to increase radiation induced death through modulation of cell cycle and reduced expression of DNA repair proteins. 10 Although treatment of cancer cells with high doses of genistein may have a growth inhibitory effect, dietary intake may result in long term exposure to much lower doses.…”
