Genetics of isozyme variants in Populus tremula, P. tremuloides and their hybrids

Abstract: Eight Populus tremula and six P . tremuloides clones as well as 49 full-sib families were studied in GOT, LAP, 6-PGDH, and SKDH by horizontal starch gel electrophoresis . For GOT one polymorphic zone was found and segregation of seven full-sib families suggests Mendelian inheritance . For LAP and 6-PGDH two zones each were clearly scored . For LAP two polymorphic loci were proposed based on the phenotypic segregation of isozyme variants in six and 34 full-sib families, respectively . In 24 full-sibs families t… Show more

“…Interpretation of MDH system is not always easy in forest species due to several coding loci and the presence of hybrid bands (THORMANN and STEPHAN, 1993). Support for the following interpretations can be found in previous research on Populus species (GALLO and GEBUREK, 1991;ALBA and AGÚNDEZ, 2000). The PGI system was revealed as monomorphic for the studied populations, hence it was not been considered for the multilocus genotypes.…”

“…Zhang et al (1992) studied P. tomentosa, a species related to P. alba, scoring two staining zones and identifying two alleles for each zone. Gallo and Geburek (1991) in P. tremula identified two loci Got-1 with tree alleles; and Got-2 with two alleles, as we have interpreted in P. alba.…”

“…Isozyme markers have been used in genetic variation studies of different Populus species (Legionet and Lefèvre, 1996), in the characterisation and differentiation of clones and species Dancik 1992a, 1992b;Castillo and Padró 1987) and in genetic studies (Müller-Starck, 1992;Gallo and Geburek, 1991).…”

“…The staining procedures and buffers followed those described by Alba & Agúndez (2000) and Wendel & Weeden (1989). An extensive review of literature on Populus species was used to interpret the results for different enzyme systems (Gallo & Geburek 1991, Rajora & Dancik 1992, Culot et al 1995, Alba & Agúndez 2000, López-de-Heredia et al 2004.…”
