Genetics of Actinorhodin Biosynthesis by Streptomyces coelicolor A3(2)

Abstract: 35 ~ ~~A series of 76 mutants of Streptomyces coelicolor A3(2) specifically blocked in the synthesis of the binaphthoquinone antibiotic actinorhodin were classified into seven phenotypic classes on the basis of antibiotic activity, accumulation of pigmented precursors or shunt products of actinorhodin biosynthesis, and cosynthesis of actinorhodin in pairwise combinations of mutants. The polarity of cosynthetic reactions, and other phenotypic properties, allowed six of the mutant classes to be arranged in the m… Show more

“…This antibiotic was produced by strains unable to synthesize any of the three antibiotics previously identified from S. coelicolor A3(2) -methylenomycin (Kirby et al, 1975;Wright & Hopwood, 1976a), actinorhodin (Wright & Hopwood, 19763;Rudd & Hopwood, 1979) or undecylprodigiosin (the 'red' antibiotic; Rudd & Hopwood, 1980; S-W. Lee, C-J. Chang & H. G. Floss, personal communication).…”

“…In spread plates discrete colonies were formed whose final diameter decreased as the concentration of the spore inoculum increased. In crowded plates mycelium at the colony margin was more sparse and less branched while growth of approaching colonies slowed down and was often associated with production of a blue pigment (Rudd & Hopwood, 1979). Occasionally, however, unrestricted margins merged to continue growth as a single colony with sporulation and pigmentation bands coinciding.…”

“…Thus, it appears that the dauG strains are dauminus regulatory mutants which may be analogous to act11 mutants of S . coelicolor (Rudd & Hopwood, 1979;Hallam et al, 1989).…”
