Genetic variations in MicroRNA genes and cancer risk: A field synopsis and meta‐analysis

Abstract: Background Cancer risk has been associated with certain gene variations in microRNA (miRNA), but conflicting evidence warrants re‐assessing of significant results in meta‐analyses. We summarized published meta‐analyses that assess the associations between miRNA polymorphism and cancers to show the validity of the findings. Method We searched PubMed and investigated the results of meta‐analyses published through November 2018. We re‐assessed the results based on false‐positive report probability (FPRP) to test … Show more

“…Even if the polymorphism does not affect the level of expression of the mature miRNA, it may reflect on the regulatory functions of miRNA on its target genes. The miRNA genes polymorphisms may modify the transcription process on several levels: firstly, on the transcription level of the primary transcript as SNPs in miRNA biogenesis genes and pri-miRNA SNPs; secondly, on the level of pri-miRNA and pre-miRNA processing as SNPs in miRNA biogenesis genes and pri-, pre-miRNA SNPs; thirdly, on the level of miRNA–mRNA interactions as SNPs in mature miRNA sequences and miRNA-binding sites [ 6 , 7 ].…”

“…MicroRNAs (miRNAs) are small non-coding RNAs of 19-25 nucleotides that can modulate gene expression by hybridizing to complementary target mRNAs, resulting in either mRNA degradation or direct inhibition of translation [4]. miRNA can also activate gene expression by interacting with complementary regions found in the promoter and coding region, as well as the 3ʹUTR of mRNA targets [5].…”

“…Firstly, our meta-analysis included data from America, Europe and Asia without Australia and Africa. Also, the sample could lose to follow-up before determining whether SCID occurred or not [25]. If we own the comprehensive data contained different continents, the result will be more representative and persuasive.…”
