Nonstructural protein 4 (NSP4), encoded by group A rotavirus genome segment 10, is a multifunctional protein and the first recognized virus-encoded enterotoxin. The NSP4 gene has been sequenced, and five distinct genetic groups have been described: genotypes A-E. NSP4 genotypes A, B, and C have been detected in humans. In this study, the NSP4-encoding gene of human rotavirus strains of different G and P genotypes collected from children between 1987 and 2003 in three cities of West Central region of Brazil was characterized. NSP4 gene of 153 rotavirus-positive fecal samples was amplified by reverse transcriptase-polymerase chain reaction and then sequenced. For phylogenetic analysis, NSP4 nucleotide sequences of these samples were compared to nucleotide sequences of reference strains available in GenBank. Two distinct NSP4 genotypes could be identified: 141 (92.2%) sequences clustered with NSP4 genotype B, and 12 sequences (7.8%) clustered with NSP4 genotype A. These results reinforce that further investigations are needed to assess the validity of NSP4 as a suitable target for epidemiologic surveillance of rotavirus infections and vaccine development.

Acute diarrheal disease is still one of the major public health problems worldwide. Rotaviruses (RV) are the most important viral etiologic agents and children under fi ve years of age are the target population. Objective: To investigate the rate of RV infection in hospitalized patients due to acute diarrhea in the cities of Ponta Grossa, Londrina and Assai -Paraná. Methods: Latex agglutination (LA); immunochromatography (ICG); polyacrylamide gel electrophoresis (PAGE) and negative staining electron microscopy (ME) tests were used to detect the virus. For the genotyping, RT-PCR and RT-PCR-ELISA were used, respectively, for NSP4 and VP4/VP7. Result: Out of 124 samples there were 69 positive stool samples for RV, for at least one of the used tests, 67 of them being RV group A (RV-A). Overall, most of the RV positive stool samples came from children under thirteen years of age. However, 12 positive cases occurred in patients aged 13 years or above, including an 81-year old patient. Conclusion: The data showed similar electropherotypes and genotypes G, P and NSP4 of the inland wild circulating strains of RV.

In an effort to detect rapidly and conveniently the infectious waterborne group A human rotaviruses that cause diarrhea in infants and children, the authors developed the first-ever cell-culture-based immunochromatography method. An immunochromatography kit interacted only with the human rotaviruses VP6 antigen, and the cell-culture-based immunochromatography detected group A human rotaviruses with a sensitivity as low as 1.99 TCID50 ml(-1). This detection sensitivity was similar to that of the cytopathic effect-based method. There were no actual differences between the sensitivity of this method and that of the real-time reverse transcription polymerase chain reaction method, which is known as a method with a relatively high sensitivity. Furthermore, while cell culture detection methods, that is total culturable virus assay, can determine only the presence of infectious waterborne viruses, the cell-culture-based immunochromatography is advantageous for the accurate detection of group A human rotaviruses. Compared with the real-time reverse transcription polymerase chain reaction method, the cell-culture-based immunochromatography is advantageous because it requires a relatively simple process that enables easy quality controls and low test costs. Thus, this study proposed a new method for the identification of group A human rotaviruses, and it is suggested that this cell-culture-based immunochromatography may be applied to detect group A human rotaviruses in aquatic environments.
