Recent genome-wide association studies have identified 14 genetic variants associated with bladder cancer in Caucasians. The effects of these risk variants and their cumulative effects in Asian populations are unknown. We genotyped these newly identified variants in a case-control study of 1,050 patients diagnosed with bladder cancer and 1,404 controls in the Chinese population. Odds rations (ORs) and 95% confidence intervals (CIs) were computed by logistic regression, and cumulative effect of risk alleles were evaluated. Overall, seven of the 14 variants were significantly associated with bladder cancer risk (p 5 9.763 3 10 23 for rs9642880 at 8q24.21, p 5 3.004 3 10 23 for rs2294008 at 8q24.3, p 5 0.012 for rs798766 at 4p16.3, p 5 0.034 for rs1495741 at 8p22, p 5 2.306 3 10 24 for GSTM1, p 5 8.507 3 10 28 for rs17674580 at 18q12.3, p 5 7.179 3 10 24 for rs10936599 at 3q26.2) and the odds ratios (ORs) ranged from 1.13 to 1.65. Moreover, there were a significant increased risk for bladder cancer positively correlated numbers of risk alleles and smoking status (P trend 5 7.060 3 10 216). However, no allelic interaction effects on bladder cancer risk were observed between cumulative effects of variants and clinical characteristics. These findings suggest that seven bladder cancer risk-associated variants (rs9642880, rs2294008, rs798766, rs1495741, GSTM1 null, rs17674580 and rs10936599) may be used, collectively, to effectively measure inherited risk for bladder cancer.Recent advances in human genomics, especially the genomewide association studies (GWAS), have improved the effectiveness of the discover of genetic variants underlying complex diseases, including bladder cancer.1 Several significant GWAS of bladder cancer have provided strong evidence for over 10 independent loci and single-nucleotide polymorphisms (SNPs) that reach the statistical genome-wide significance. Background: Cytochrome P450 2E1 (CYP2E1) might be involved in the development of bladder cancer. However, previous studies of any association between CYP2E1 RsaI/PstI polymorphism and bladder cancer risk have yielded conflicting results. In this study, we performed a more precise estimation of the relationship by a meta-analysis based on the currently available evidence from the literature. Method: To assess the effect of CYP2E1 RsaI/PstI polymorphism on bladder cancer susceptibility, a meta-analysis of 6 available studies with 1,510 cases and 1,560 controls were performed through Feb 2014. Summary odds ratios (ORs) and corresponding 95% confidence intervals (CIs) were used to estimate the strength of association for CYP2E1 RsaI/PstI polymorphism under different genetic models. Results: When available studies were pooled into the meta-analysis, we found that the C1C2 and C2C2 genotypes of CYP2E1 RsaI/PstI polymorphism significantly decreased bladder cancer risk

Context-Continuing advances in genotyping technologies and the inclusion of DNA collection in observational studies have resulted in an increasing number of genetic association studies.Objective-To evaluate the overall progress and contribution of candidate gene association studies to current understanding of the genetic susceptibility to cancer.Data Sources-We systematically examined the results of meta-and pooled analyses for genetic polymorphisms and cancer risk published through March 2008.Study Selection-We identified 161 meta-and pooled analyses, encompassing 18 cancer sites and 99 genes. Analyses had to meet the following criteria: 1) at least 500 cases, 2) cancer risk as outcome, 3) not focused on HLA genetic markers, and 4) published in English.Data Extraction-Information on cancer site, gene name, variant, point estimate and 95% confidence interval, allelic frequency, number of studies and cases, tests of study heterogeneity and publication bias were extracted by one investigator and reviewed by other investigators.Results-These 161 analyses evaluated 344 gene-variant/cancer associations and included on average 7.3 studies and 3,551 cases (range: 508-19,729 cases) per investigated association.
