Tigano-Milani (1995) proposed the hypothesis that more than one haplotype (clone) may be required to initiate and maintain an epizootic in a natural and heterogeneus insect population, such as the one found under Colombian conditions. “…Genomic DNA was extracted using a universal rapid salt method (Aljanabi & Martinez, 1997). The PCR reactions were performed in 30-µL volume, with 15 ng of each template, using the PTC-100 programmable thermal controller, and a temperature profile already described (Tigano-Milani et al, 1995). The amplifications were done using the following reaction mix: 1 unit of Taq polymerase, 1x Taq polymerase reaction buffer, 4 mM of MgCl 2 , 200 µM of each deoxynucleotides triphosphate, and 1 µM of 10-mer primer.…”
