Genetic risk factors for ME/CFS identified using combinatorial analysis

Abstract: Background Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a debilitating chronic disease that lacks known pathogenesis, distinctive diagnostic criteria, and effective treatment options. Understanding the genetic (and other) risk factors associated with the disease would begin to help to alleviate some of these issues for patients. Methods We applied both GWAS and the PrecisionLife combinatorial analytics platform to analyze ME/CFS c… Show more

“…Moreover, various environmental triggers, including infectious agents, toxins, and physical or emotional stressors, have been proposed as potential triggers that can induce or exacerbate mitochondrial dysfunction in susceptible individuals. These triggers may lead to mitochondrial damage, oxidative stress, and inflammation, further impairing mitochondrial function and perpetuating the cycle of fatigue and other core symptoms in people with PVFS [ 50 , 51 , 52 , 53 , 54 , 55 ].…”

“…However, very recently, compelling support for genetic susceptibility came from a combinatorial analysis of clusters of loci from a genome-wide association study. It identified clusters of 199 single-nucleotide polymorphisms connected to 14 genes that could identify 91% of patients from the UK biobank selected for the study [ 27 ]. These genes could be linked to processes likely dysfunctional in ME/CFS.…”

“…These disease signatures capture both the linear and non-linear effects of genetic and molecular interaction networks and enable the identification of associations including those that are only relevant to a subgroup of patients. We have previously validated this analytical approach across a variety of complex chronic diseases where it has identified more associations with increased explanation of observed disease variance and reproducibility than comparable GWAS studies 15,16,17 .…”
