Genetic relationship between acylpeptide hydrolase and acylase, two hydrolytic enzymes with similar binding but different catalytic specificities.

Abstract: An 87% identity has been found between the reported cDNA sequence that encodes acylpeptide hydrolase (EC 3.4.19.1) [Mitta, M., Asada, K., Uchimura, Y., Kimizuka, F., Kato, I., Sakiyama, F. & Tsunasawa, S. (1989) J. Biochem. 106, 548-551] and a cDNA transcribed from a locus (DNFISS2) on the short arm ofhuman chromosome 3, reported by Naylor et al. [Naylor, S. L., Marshall, A., Hensel, C., Martinez, P. F., Holley, B. & Sakaguchi, A. Y. (1989) Genomics 4,[355][356][357][358][359][360][361]; the DNF15S2 locus … Show more

“…This report demonstrates DPP IV activity in I100. The PSM/brain NAALADase activity is reminiscent of that described for acylaminoacyl peptidase (50,51). PSM seems to have dual specificities, as it has been shown to also have pteroyl poly-␥-glutamyl carboxypeptidase activities (32,33).…”

“…The Acy1 gene has been assigned to chromosome 3p21·1; this region is reduced to homozygosity in small-cell lung and renal cancer (Miller et al 1989). Accordingly, aminoacylase 1 has been reported to exhibit reduced or absent expression levels in SCLC cell lines and various tumours (Jones et al 1991, Scaloni et al 1992. These findings should support the hypothetical role of p53 in Acy1 gene expression control.…”

“…In eukaryotic cells, a strong degree of genetic similarity between AARE and aminoacylase was suggested by Jones et al (33). In P. horikoshii, however, the gene for aminoacylase was found at another locus in the genome (27), and AAREP did not share homology or activity with aminoacylase.…”
