Genetic profile of Mycobacterium tuberculosis and treatment outcomes in human pulmonary tuberculosis in Tanzania

Abstract: Information on the different spoligotype families of Mycobacterium tuberculosis in Tanzania is limited, and where available, restricted to small geographical areas. This article describes the genetic profile of M. tuberculosis across Tanzania and suggests how spoligotype families might affect drug resistance and treatment outcomes for smear positive pulmonary tuberculosis patients in Tanzania. We conducted the study from 2006 to 2008, and the isolates were obtained from samples collected under the routine drug… Show more

“…Similar families have been reported in variable proportions in other countries bordering Tanzania [31e35], indicating a geographical widespread of this group of spoligotypes in the region. These findings are also in agreement with those of other studies conducted in Tanzania which reported the predominance of these families [21,36,37].…”

“…Strain families previously reported in Tanzania but not reported in this study include the Haarlem genotype [21,37] and the X genotypes [21,36e38]. We detected a high proportion of T family isolates as found in previous studies [21,37].…”

“…NTM infection does not respond to anti-TB agents; however, the clinical impact of the indiscriminate use of anti-TB drugs in the tremendously, recently emerging drug-resistant TB era remains unassessed 2,24. Infection due to species of M. fortuitum and M. abscessus , which both have the capacity to cause respiratory infections in human, has previously been reported in Tanzania 25,26. Although this study did not conduct drug sensitivity testing, both M. fortuitum and M. abscessus can be misdiagnosed as resistant TB/multidrug-resistant TB due to their natural resistant pattern to antituberculosis drugs,15,26 which may result in unnecessary administration of toxic second-line anti-TB treatment.…”

“…These previous studies are limited as they only focused on few geographical locations and used spacer oligonucleotide typing (spoligotyping) technique which has limitations for phylogenetically robust strain classification [22,23]. Only one study profiled MTBC on a countrywide scale albeit with low sampling coverage [20].…”
