Genetic predisposition for chronic venous insufficiency in several genes for matrix metalloproteinases (MMP‐2, MMP‐9, MMP‐12) and their inhibitor TIMP‐2

Abstract: Studying these polymorphisms can contribute to better identification of patients at higher risk of developing CVD, while providing the most appropriate prevention and treatment strategies for limiting the progression and complications of CVD.

“…Although a number of genes have been implicated, 89 to date, no causal association has been proved. 90 GWASs in >400 000 individuals established 12 candidate loci for varicose veins in individuals with European ancestry, highlighting the SNPs CASZ1 , PIEZO1 , PPP3R1 , EBF1 , STIM2 , HFE , GATA2 , NFATC2 , and SOX9 .…”

“…More recently, it has been shown (7,18) that specific polymorphisms in the MMP-9 and TIMP-2 genes potentially increase the risk of developing varicose veins in patients. Vein remodeling and associated gene expression were also studied, but the results did not correlate with the development and severity of PTS [5,12]. However, their preliminary data showed an increase in MMP-9 expression and a decrease in Toll-like receptor 9 expression in acute deep vein thrombosis.…”

“…Both MMP and TIMP regulate extracellular matrix homeostasis. MMPs are involved in the degradation of extracellular matrix components, while TIMPs affect vascular remodeling by causing changes in venous elasticity [6,12]. MMPs can also influence biologically active molecules present on the cell surface through G protein-coupled receptors (GPCRs) and regulate the cellular environment [13].…”

“…We must emphasize that after the visit of a Vascular Specialist the prescription of venoactive drugs and graduated compression stockings rose 3-fold. This clearly shows the importance of being taken in charge by a vascular specialist (6, [26][27][28][29][30].…”
