Genetic polymorphism of miRNA-196a and its target gene annexin-A1 expression based on ethnicity in Pakistani female breast cancer patients

Abstract: Objective: To evaluate the association of miR-196a rs11614913 C/T genetic variation and its target gene annexin A1 mRNA expression with breast cancer risk in Pakistani female ethnicities. Methods: This case control study, conducted from March 2017 to November 2018 included 295 breast cancer patients, 295 controls of three Pakistani ethnicities and archived 100 samples of cohort group for genotyping and expression profiling. Genotyping of miR-196a (rs11614913 C/T) was done by ARMS PCR technique. Annexin-A… Show more

“…14,15 It can reach tumor cells in the vascular endothelial space and undergo a liquid-to-gas phase transition to enhance the ultrasound imaging, so it has been widely used in ultrasound molecular imaging. [16][17][18][19] You et al 20 Prepared a multifunctional Fe3O4-PFH/PLGA nanoparticle, which undergoes a phase change in the PFH core of highly focused ultrasound and can work in synergy with highintensity focused ultrasound to effectively improve the therapeutic effect of tumors. In this study, in vitro targeting experiments showed that many nanoparticles labeled with red fluorescence in the AP-GNP group were tightly attached to the cell membrane surface of MCF-7 labeled with green fluorescence, and some nanoparticles were swallowed.…”

“…Furthermore, one of the first discovered miRNA, miRNA-196a, which is upregulated in the vast majority of cancer types including NSCLC, was shown to be involved in the mediation of cisplatin resistance; however, its mechanism is not clear [75]. MiRNA-196a targets the Annexin-A1 (ANXA1) gene that regulates physiological mechanisms such as hormone secretion, apoptosis, exocytosis, and signal transduction [76]. ANXA1 is also involved in the acquisition and maintenance of a cancer stem cell-like phenotype that is characterised by upregulation of several chemoresistant mechanisms, including the activity of ABC proteins [77,78].…”

“…ANXA1 plays a significant role in membrane trafficking, exocytosis, signal transduction, cell differentiation, and apoptosis, among other biological roles ( Luthra et al, 2008 ). Overexpression of miR-196a2 due to variant rs11614913 leads to the suppression of ANXA1 thereby promoting cell proliferation and suppressing apoptosis ( Solito et al, 2001 ; Luthra et al, 2008 ; Rahim et al, 2019 ). ANXA1 exhibits varied expression in different cancers.…”

“…ANXA1 exhibits varied expression in different cancers. It is upregulated in glioma and oropharyngeal cancer and downregulated in prostate cancer, esophageal squamous cell, and head and neck squamous carcinoma ( Rahim et al, 2019 ). As an upstream regulator, miR-196a has been found to partially direct the cleavage of the mRNA of the HOX gene clusters.…”
