Genetic polymorphism at codon 10 of the transforming growth factor-β1 gene in patients with alcoholic liver cirrhosis

Abstract: Background/AimsTransforming growth factor beta1 (TGF-β1) is a key cytokine in the production of extracellular matrix. A genetic polymorphism at codon 10 of the TGF-β1 gene is associated with liver fibrosis. We investigated the effect of genetic polymorphisms at codon 10 on the development of alcoholic liver cirrhosis (ALC).MethodsIn total, 119 controls and 182 patients with ALC, were enrolled in the study. Clinical and laboratory data including total lifetime alcohol intake were collected at enrollment. The ge… Show more

“…Single-nucleotide polymorphisms associated with increased risk of ALD include the −238AA TNF- α variant [odds ratio (OR) 2.36, 95% CI 1.1–5.3], −511C IL-1 β allele (OR 2.64, 95% CI 1.2–5.9), −509C TGF- β 1 allele (OR 4.71, 95% CI 1.7–13.1), and −592A IL-10 allele (OR 2.17, 95% CI 1.1–4.8) [ 26 ]. There were no differences between genetic polymorphism at codon 10 in the TGF- β gene and the development of alcoholic liver cirrhosis in a Korean population [ 27 ].…”

“…Abdominal magnetic resonance imaging (MRI) or USG guided liver biopsy were performed on a separate day if needed diagnostically. A heavy alcohol consumer was defined as a person who consumed more than 80 g of alcohol per drinking session and drank more than 5 times a week for more than 10 yr (9). All USG, CT and MRI were assessed by radiologists.…”

“…Genetic influences have been implicated to play a significant role in the development of liver disease [ 9 - 10 ]. One such example of a genetic influence on liver disease is the hereditary hemochromatosis gene, which causes the body to absorb too much iron from food [ 11 ].…”

“…The transforming growth factor-beta 1 (TGF-β1) is a cytokine that plays an important role in the pathogenesis of liver disease [ 9 - 10 ]. Several studies have investigated the association between genetic variations in TGF-β1 and the levels of TGF-β1 with the development and progression of liver disease [ 9 - 10 , 14 - 15 ].…”
