Genetic or pharmacological superoxide-hydrogen peroxide imbalances modulate the in vitro effects of lithium on glycogen synthase kinase-3β

Abstract: as expected PHA exposure triggered a strong upregulation of GSK-3β gene expression (p ≤ 0.001), and Li exposure showed GSK-3β gene downregulation from 0.7 mEq/L concentrations. However, Li modulatory effects on GSk-3β gene and protein expression was directly influenced by basal S-HP balance. Presence of high S-basal levels (VV genotype and VV-like cells) induced attenuated Li anti-inflammatory effects in comparison with balanced and AA and AA-like cells (p < 0.001). Despite methodological limitations related t… Show more

“…Likewise, açai was able to reverse the inflammatory activation of macrophages [49] and microglia [50]. Other studies have also shown that the inflammatory pattern associated with the VV-SOD2 genotype was attenuated in vitro by supplementing cell cultures with lithium [51]. On the other hand, genetic variations leading to mitochondrial dysfunction could also contribute to the risk of mood disorders [52 -54].…”

“…The range evaluated for OLZ was 0.02 μg/mL to 0.08 μg/mL according to guidelines of drug therapeutic monitoring [39]. The therapeutic Li concentration used was 0.07 mEq/L, which has been previously evaluated in vitro [21,22]. Therefore, results of this study may be representative of interactive in vivo effects between OLZ and Li.…”

“…All protocols performed were similar to previously described studies by Barbisan et al [22] and Duarte et al [23] of in vitro psychotropic drug effects on macrophage inflammatory response. Macrophages are highly sensitive to antigens and other antigenic factors, which induce intense cellular proliferation and morphological changes.…”

“…Similarly, low brain Nrf2 expression occurs in hypogonadal 21-month-old male rats, an effect countered by low dose T administration (Zhang et al, 2016;Cui et al, 2017), while chemical castration of rats lowers testicular Nrf2 protein expression (Bae et al, 2017). Because excess oxidative stress itself increases GSK3β protein expression (Shin et al, 2006;Barbisan et al, 2018), excess oxidative stress may act in a feed-forward manner to increase GSK3β activity, inhibit Nrf2 activity, and further increase oxidative stress. GSK3β also upregulates Aβ production (Ryder et al, 2003) and it increases tau hyperphosphorylation (Papasozomenos, 1997;Papasozomenos and Shanavas, 2002).…”
