Pea Fusarium wilt, incited by Fusarium oxysporum f. sp. pisi (Fop), has always been a devastating disease that causes severe yield losses and economic damage in pea-growing regions worldwide. The utilization of pea cultivars carrying resistance gene is the most efficient approach for managing this disease. In order to finely map resistance gene, a F2 population was established through the cross between Shijiadacaiwan 1 (resistant) and Y4 (susceptible). The resistance genetic analysis indicated that the Fop resistance in Shijiadacaiwan 1 was governed by a single dominant gene, named as FwS1. Based on the bulked segregant analysis sequencing (BSA-seq) analyses, the gene FwS1 was initially detected on chromosome 6 (i.e., linking group Ⅱ, chr6LG2), and subsequent linkage mapping with 589 F2 individuals fine-mapped the gene FwS1 into a 91.4 kb region. The further functional annotation and haplotype analysis confirmed that the gene Psat6g003960, characterized by a NB-ARC (nucleotide-binding adaptor shared by APAF-1, R proteins, and CED-4) domain, was considered as the most promising candidate gene. The encoding amino acids were altered by a “T/C” single-nucleotide polymorphism (SNP) in the first exon of the Psat6g003960, resulting in the observed differences of Fop resistance in peas. Based on this SNP locus, the molecular marker A016180 was determined to be a diagnostic marker for FwS1 by validating its specificity in both pea accessions and genetic populations with different genetic backgrounds. The FwS1 with diagnostic KASP marker A016180 could facilitate marker-assisted selection in resistance pea breeding in pea. In addition, upon comparing the candidate gene Psat6g003960 of 74SN3B and SJ1, it was noted that their sequences are identical, suggesting that the FwS1 and Fwf may be the same resistance gene against Fusarium wilt.

Cereal production is of strategic importance to the world economy. Although the primary aim of breeding programs is to develop cultivars with improved agronomic performance, including high grain yield and grain quality, as well as disease and lodging resistance, nowadays the adaptability to changing environmental conditions seems to be an extremely important feature. The achievement of these breeding objectives in diploid cereal species such as rice, barley, or maize is straightforward. The genetic improvement of polyploid crops such as hexaploid wheat and oats for increased crop production is highly demanding. Progenitor species and wild relatives, including taxa at lower ploidy levels, have preserved a high degree of useful genetic variation. The world’s genebank collections of wheat and oat germplasm provide extremely rich resources for future breeding and utilization. This review highlights the immense potential of cultivated wild relatives as donors of genes for a wide range of biotic and abiotic traits and their impact on wheat and oat breeding. This review covers methods allowing access to these genetic resources, and it highlights the most (and most recently)-exploited related species for gene introgression in wheat and oats. Further, it will also deal with the impact of genomics and cloned genes on the advanced discovery, characterization, and utilization of genetic resources in these two cereals.

Wheat powdery mildew is a fungal disorder caused by Blumeria graminis f. sp. tritici (Bgt) and is a severe and significant threat to the yield and quality of its host. The most practical and environmentally friendly approach to controlling this disease is through resistance gene identification to develop resistant varieties. Wild germplasm relatives of wheat are a valuable reservoir of genes contributing to resistance against powdery mildew. In our study, we identified the Aegilops tauschii germplasm “KU-2013”, exhibiting seedling resistance to Bgt isolate E09 following hexaploidization. Genetic analysis and chromosomal localization of the powdery mildew resistance gene in doubled haploid (DH) KU-2013 indicated that the disease resistance gene in DHKU-2013 is governed by a dominant gene situated in 5DS, tentatively named PmKu-2013. Following the analysis of PmKu-2013 relative to the genes at the Pm2 locus, it was inferred that PmKu-2013 represented a distinct novel gene separate from Pm2. Using molecular marker analysis, PmKu-2013 was found to be ultimately mapped between the sdau5DS5-3 and sdau5DS6-1 markers, with genetic distances of 0.6 cM and 1.3 cM, respectively. Using markers tightly linked to PmKu-2013, the genotypes of core wheat varieties from various regions were identified, laying the foundation for the transfer and utilization of PmKu-2013 in molecular-assisted selection (MAS) for breeding.
