Genetic Heterogeneity and Recombination in Human Type 2 Astroviruses

Abstract: dNovel lineages of human astrovirus (HAstV) types 2, 2c, and 2d have been identified. Upon sequencing of the 3= end of the genome, the type 2c and 2d HAstVs were found to be open reading frame 1b (ORF1b)-ORF2 recombinant, with ORF1b being derived from type 3 and type 1 HAstVs, respectively. An ORF2 interlineage recombinant strain, 2c/2b, was also identified.

“…Recombinations between different animal AstVs have also been described (152,155,162,183). Although most recombination breakpoints have been identified upstream of the conserved ORF1b/ORF2 junction region (155,168,172,182), they are also frequent within ORF2 (152,155,172,177), and they may also exist within ORF1a (155,181) and ORF1b (155,177,180).…”

“…As previously reported, the ORF1b/ORF2 region was known as a crossover point [ 22 24 ]. There were several appearance of recombination event occurred in this region from different lineages or among same serotype [ 23 26 ]. Unfortunately, we failed to amplify the whole genome of CAstVs, as it was hard to evaluate the actual origin or recombination with other species.…”

“…So far, there are over 80 avian and mammalian host species detected in many species, which leads to its large diversity [ 27 ]. Generally, the majority of AstVs naturally infect only related host species, or more than one lineage could transmit in a single host [ 23 , 28 ]. Whether this recombination event or cross-species transmission will emerge in CAstVs should raise corncens.…”

“…1). Virus recombination has also been reported to drive the increasing genomic diversity of this virus family (3,4), which may arise from infection of individuals with multiple virus strains belonging to different lineages, although reporting of multiple infections is poor. Systematic astrovirus surveillance in swine in the United States revealed a high proportion of pigs (13.9%) coinfected with viruses from two or more porcine astrovirus lineages (5), signifying the potential for recombination.…”
