Genetic Evidence for Adenylyl Cyclase 1 as a Target for Preventing Neuronal Excitotoxicity Mediated by N-Methyl-D-aspartate Receptors

Abstract: The excessive activation of N-methyl-D-aspartate (NMDA) receptors by glutamate results in neuronal excitotoxicity. cAMP is a key second messenger and contributes to NMDA receptordependent synaptic plasticity. Adenylyl cyclases 1 (AC1) and 8 (AC8) are the two major calcium-stimulated ACs in the central nervous system. Previous studies demonstrate AC1 and AC8 play important roles in synaptic plasticity, memory, and persistent pain. However, little is known about the possible roles of these two ACs in glutamate-i… Show more

“…Bilateral ACCs from the mice were dissected and homogenized in lysis buffer containing 10 mM Tris-HCl, pH 7.4, 2 mM EDTA, 1% SDS, 1ϫ protease inhibitor mixture (Sigma), and 1ϫ phosphatase inhibitor mixture I and II (Sigma). The analysis was performed according to the procedures as reported previously (Wang et al, 2007). Briefly, the protein samples were quantified by Bradford assay.…”

“…Although the molecular mechanism for NB001 is unclear, it still has important analgesic effects in animals (Wang et al, 2011). AC1 has roles in both pain sensation (Vadakkan et al, 2006;Wang et al, 2007) and memory (Villacres et al, 1998). The ability of NB001 to impair pain sensation, but not memory capability, is consistent with an indirect effect on AC1.…”
